Objective:Research Zhong feng gao preadaptation stimulates the brain ischemia Tolerance(Ischemic how IT),to produce endogenous protective substances to exert delayed cerebral ischemia and reperfusion injury,and to explore the protection mechanism,provide experimental support for the study of the prevention of Ischemic stroke.Methods:SPF came SD rats,weight280±20g,were randomly divided into normal group(K)and control group(J),cerebral ischemia reperfusion group(IR)and cerebral ischemia preconditioning group delay protection model 3 days(M3),delayed cerebral ischemia preadaptation protection model group 5 days(M5),ischemic preconditioning group 7 days delay protection model(M7),Zhongfenggao preconditioning group(Z3)three days delay protection,Zhongfenggao preadaptation 5 days delay protection group(Z5),stroke,paste in 7days delay protection group(Z7).The normal feeding of K group is same as that of other groups.J group only exposed left CCA,ECA and ICA,but did not block MCA;IR group established rats Cerebral ischemia reperfusion(CIRI)model based on Longa method.M group established the Ischemic preconditioning(IPC)model according to Longa method.Group Z:pretreatment of apoplexy with suspension and CIRI model.The rats were observed in the experiment.After anesthesia,the rats were examined for neurological impairment score.In addition to group K and J group,CIRI24 h of the remaining group was severed head,and the brain tissue HE staining was observed.The activity of NO and NOS in cerebral cortex was measured by colorimetry.ELISA was used to detect cortical protein kinase C(PKC)and heat shock protein(HSP70)concentration.The immunohistochemical method was used to detect the expression of NOS,PKC and HSP70 in the hippocampus and the number of positive neurons.Rt-pcr was used to detect the expression of NOSmRNA,PKCmRNA and HSP70 mRNA in the cerebral cortex.Results:(1)Preconditioning of apoplexy cream can help cerebral ischemia rats recover in general,indicating that preconditioning of apoplexy cream can reduce the effects of cerebral ischemia on diet and mental weight.(2)Apoplexy ointment can reduce neurologic function score,indicating that apoplexy ointment can reduce cerebral ischemic nerve function damage.(3)The observation of brain tissue sections showed that apoplectin can reduce apoptosis caused by cerebral ischemia and changes in brain structure,indicating that apoplectin preconditioning can protect brain tissue.(4)Stroke paste by adding protective material in brain tissue PKC and HSP70 protein and gene expression,cell apoptosis neurotransmitter NO reduction and its speed limit NOS enzyme activity and NOS protein and gene expression,exert its preadaptation delay protection,its mechanism is to reduce brain cells,peroxide stability balance of Ca2 +,inhibiting cell apoptosis and the release of inflammatory mediators,protect the brain tissue.Conclusion:Preconditioning of stroke cream can nondestructive produce IPC delayed protection effect,induce Ischemic Tolerance(IT),and reduce CIRI.Its mechanism may be the IPC effect inhibition of Ischemic cerebrovascular disease(Ischemic cerebrovascular diseases,ICVD)caused by cell apoptosis,stable Ca2 + channel,reduce ischemia irritability peroxidation caused by injury,inhibit inflammation ischemia.This study provides a theoretical basis for the use of apoplexy ointment in clinical preconditioning to produce delayed protective effect and reduce cerebral ischemia damage and reperfusion injury.
