Effects Of Electroacupuncture Combined With Brain Injection VEGF On Endoplasmic Reticulum Stress Related Proteins In Rats With Cerebral Ischemia-reperfusion Injury

ObjectiveTo observe the effect of electroacupuncture and electroacupuncture combined with intracerebral injection of VEGF on endoplasmic reticulum stress(ERS)related protein ATF6,IRE1,CHOP,XBP1,GRP78 of cerebral ischemia reperfusion injury(CIRI)rats and study the repair effect of electroacupuncture combined with intracerebral injection of VEGF on CIRI.MethodsA total of 68 male SD rats were randomly divided into sham operation group(J),model group(M),electroacupuncture group(EA),brain injection VEGF group(VEGF),electroacupuncture combined with brain injection VEGF group(EA+VEGF).including 8 in the J group and 15 in the rest of each group.Making model for M group,EA group,VEGF group and EA+VEGF group by using thread embolism method.Rats in the J group were not be inserted the thread,others were the same as those in the M group.EA group and EA+VEGF group rats were treated with acupuncture "Baihui",left "Qu Chi" and "Zusanli" after 24 h of the model.The treatment was treated with electroacupuncture,once a day and 30 minutes each time for 14 days.At the same time,the M group,J group and VEGF group were fixed at the same time,but not acupuncture.The rats in VEGF and EA+VEGF group were injected with 10?l VEGF(0.025?g/?l)in the lateral ventricle 24 hours after modeling.After 14 days of intervention,the nerve function score(mNSS)of rats in each group was detected,the brain tissue of CIRI region of rats was observed by Nissl staining,and the content of ATF6,IRE1,XBP1,CHOP and GRP78 protein in rats was measured by Western blot method.The content of ATF6,IRE1,XBP1,CHOP,GRP78 mRNA were detected using RT-PCR.ResultsThe mNSS score in the model group was significantly higher than that in the J group(P<0.05),and the mNSS score in the electroacupuncture group,the VEGF group and the electroacupuncture group was significantly lower than that in the model group(P<0.05),and the mNSS score in the electroacupuncture group of +VEGF group was significantly lower than that in the electroacupuncture group and the VEGF group(P<0.05).Compared with the J group,the number of Nissl corpuscles in the model group was lower than that of the J group(p<0.05).Compared with the model group,the number of Nissl corpuscles in the electroacupuncture group,the VEGF group and the electroacupuncture +VEGF group were higher than that of the model group(P<0.05),and the number of Nissl corpuscles in the +VEGF group of electroacupuncture was significantly higher than that in the electroacupuncture group and the VEGF group(P<0.05).The content of ATF6,IRE1,XBP1,CHOP and GRP78 in the model group was higher than that in J group(P<0.05),and the content of ATF6,IRE1,XBP1 and CHOP protein in the electroacupuncture group,the VEGF group and the electroacupuncture +VEGF group were significantly lower than that of the model group.Compared with the electroacupuncture group and the VEGF group,the content of ATF6,IRE1,XBP1 and CHOP protein decreased significantly(P<0.05),and the content of GRP78 protein increased significantly(P<0.05).The content of ATF6,IRE1,XBP1,CHOP and GRP78 mRNA in the model group was higher than that in the J group(P<0.05),and the content of ATF6,IRE1,XBP1,and +VEGF in the electroacupuncture group,VEGF group and electroacupuncture group was lower than that in the model group.The contents of ATF6,IRE1,XBP1 and CHOP mRNA decreased compared with those in EA group and VEGF group(P<0.05),and GRP78 mRNA content increased(P<0.05).ConclusionElectroacupuncture combined with intracerebral injection of VEGF could promote tissue repair of CIRI and its possible mechanism is related to down-regulation of the expression of ERS related protein ATF6,IRE1,CHOP,XBP1 and up-regulation of the expression of ERS related protein GPR78.The effect of electroacupuncture combined with intracerebral injection of VEGF method is superior to that of single use of electroacupuncture or intracerebral injection of VEGF.
