Objective: In recent years,more and more attention has been paid to the expression of post transcriptional regulatory genes of micro RNA in various tumors.It has become a research hotspot in the pathogenesis and diagnosis of pituitary adenoma.In this experimental study we test the expression of miR-493 and Let-7b in invasive pituitary adenoma tissues and peripheral blood,and analysis its clinical significance with clinical feature,and to further explore the relationship between miR-493 and Gal-3 protein and Let-7b protein and HMGA2 in pituitary adenoma.We want to provide a theoretical basis for reference in pathogenesis and early diagnosis,targeted therapy of invasive pituitary adenoma.Methods: From June 2016 to September 2017,48 patients with pituitary adenoma admitted to the Affiliated Hospital of Qingdao University for surgical treatment in neurosurgery were included into our study.We collect intraoperative pituitary adenoma tissues and preoperative peripheral blood 3ml,and the patients of this research do not accept drugs,surgery,radiotherapy and other treatments.The pituitary tumor is divided into invasive pituitary tumor group and non invasive pituitary tumor group,according to imaging examination and intraoperative findings.We tested the relative expression of miRNA-493,Let-7b genes by real-time quantitative PCR(RT-PCR),and we tested the expression of Gal-3 and HMGA2 in pituitary adenoma by Western blot and to evaluate their correlations with miRNA-493,Let-7b.Results: 1.The expression of miR-493 was decreased in invasive pituitary adenoma tissues(4.19±0.82)relative to non-invasive pituitary adenoma tissues(5.92±0.89)(P<0.05);The expression of Let-7b was decreased in invasive pituitary adenoma tissues(3.31±0.55)relative to non-invasive pituitary adenoma tissues(4.91±0.49)(P<0.05).3.The expression levels of mi R-493 and Let-7b in invasive pituitary adenomas were lower than those in non invasive pituitary adenomas(2.65 + 0.43,2.97 + 0.21,P=0.075,1.86 + 0.43,2.07 + 0.28,P=0.080).4.The expression of miR-493,Let-7b correlated with the invasiveness of pituitary adenoma.4.It showed that there were negativerelationships between miR-493 and Gal-3(r=-0.38,P<0.05),and there were negativerelationships between Let-7b and HMGA2(r=-0.67,P<0.05).Conclusions: 1.MiR-493 may play the role of tumor suppressor gene in the occurrence and development of pituitary adenoma through Gal-3 pathway.2.Let-7b may play the role of tumor suppressor gene in the occurrence and development of pituitary adenoma through HMGA2 pathway.3.The expression of Gal-3 and HMGA2 may have important reference value in the diagnosis of invasive pituitary adenoma.
