Objective:The effects of high fructose diet on subgeneration glucose metabolism and lipid metabolism were compared,and the effect of high fructose diet on the insulin resistance in pregnancy was preliminarily investigated.Method:1.Wistar pregnant rats of 30 9 weeks old were randomly divided into 5 groups according to the body mass: blank control group(blank control,BC),2g/kg fructose group,6g/kg fructose group,18g/kg fructose group,18g/kg sucrose group,each group of 6 each,daily gavage of fructose to delivery to the stomach.Body weight,tail blood collection and fasting glucose were measured every week.The offspring of each group were randomly selected to feed 9 basic feeds,and the body weight and blood sugar were measured every week until the end of eighth weeks.2.The serum was separated and the content of fructose in serum was detected.3.The oral glucose tolerance test(OGTT)was conducted at the end of the eighth week to assess the glucose tolerance.4.The level of fasting insulin was measured,the insulin resistance index was calculated,and the function of islet was evaluated.5.The level of triglyceride and total cholesterol was detected by enzyme linked immunosorbent assay,and the level of lipid metabolism was evaluated.6.The liver and pancreas tissue were observed and weighed,and the changes were observed by hematoxylin and eosin staining.Result:1.Birth weight of the offsprings:the birth weight of the offspring of the offspring of the offspring was different.The birth weight of the 6g/kg fructose intake group was lower than that of the control group(P <0.05).The birth weight of the 18g/kg fructose intake group was compared with the birth weight P <0.05)of the control group.2.The level of serum fructose in young rats:the level of fructose in the serum of 6g/kg fructose group and 18g/kg sucrose group was significantly higher than the level of fructose in the control group(P<0.05).The level of fructose in 6g/kg fructose group and 18g/kg sucrose group was significantly higher than that of 2g/kg fructose level(P <0.05).The fructose level of 18g/kg fructose group increased significantly compared with the other four groups,and the difference was statistically significant.Meaning(P <0.05).3.Fasting blood glucose in the offspring: there was no significant difference in fasting blood glucose between the 6g/kg fructose group and the 18g/kg sucrose group compared with the control group and the 2g/kg fructose group(P >0.05).Compared with the control group,the fasting blood glucose level of the 18g/kg fructose group was higher than that of the control group,and the difference was statistically significant.4.The result of OGTT:the offspring of different doses of fructose intake group underwent OGTT test in eighth weeks.The peak blood sugar appeared at 30 min,at this time,there was no significant difference between the control group and the 2g/kg fructose group.The blood sugar values in the other groups were significantly higher than those of the two groups(P <0.05).After 30 min,the blood sugar values were all decreased in varying degrees.At 120 min,the control group and 2g/kg blood glucose value were basically recovered to the initial blood glucose value,but the rest were the rest.Each group was still higher than the initial blood sugar(P <0.05).5.The serum insulin level: the serum insulin level of 6g/kg fructose group and 18g/kg fructose group was significantly higher than that of the control group(3.87,5.97)u IU/m L,higher than 2g/kg fructose group(2.29,4.39)u IU/m L,the difference was statistically significant(P <0.05),6g/kg fructose group,18g/kg fructose group,18g/kg sucrose group HOMA-IR value was significantly higher than the control group,2g/kg.Fructose group,the difference was statistically significant(P <0.05).6.Blood lipid in the offspring:the levels of triglyceride(TG)and total cholesterol(TC)in each group were different.Compared with the control group and the 2g/kg fructose group,the triglyceride and total cholesterol in the 18g/kg fructose group were significantly higher than those in the 2g/kg fructose group,and the difference was statistically significant(P <0.05).7.Viscera index of rat liver:the hepatic coefficient of the offspring of the 6g/kg fructose group offspring and the 18g/kg fructose group was 1.26-fold and 1.39-fold higher than that of the blank control group,and the difference was statistically significant(P <0.05).8.Morphological changes of the liver:the liver lobules in the control group and the 2g/kg fructose group were normal and polymorphic.The nuclei were complete and round or oval,and the demarcation between the cells was observed and the cytoplasm was uniform.The 6g/kg fructose group had only a small amount of fat droplets in the group of fructose groups,and a large number of fat droplets gathered under the 18g/kg fruit sugar group,showing fat.It is said denaturation?9.Morphological changes of the pancreas:in the control group and 2g/kg fructose group,the islets were located in the center,spherical or oval,and the cells were filled and swayed.The islets in other groups were different in location,abnormal in morphology and unclear in islet border,especially in 18g/kg fructose group.Conclusion:1.High and super high fructose intake during pregnancy will lead to impaired glucose tolerance and abnormal lipid metabolism in offspring.2.Heavy fructose intake during pregnancy can lead to adverse pregnancy outcomes.3.High-dose and ultra-high fructose intake during pregnancy increases the risk of insulin resistance in offspring.4.Pregnant women should eat fruits reasonably(200-350g).
