| Objective: The purpose of this study was to compare the expression of the marker of tumor environment and cencer stem cell(CSC)in the serum of the patients with non-small cell lung cancer(NSCLC)such as vascular endothelial growth factor receptor 1(VEGFR1),aldehyde dehydrogenase 1 a1(ALDH1A1),fibroblast activation protein(FAP),platelet-derived growth factor AA(PDGF-AA)with healthy control,and clarify the relationship between cytokines and NSCLC clinical pathological features,explore the significance of the environment and CSC in occurrence,development,invasion and metastasis of NSCLC.Methods: Collected 54 serum sample of NSCLC without any treatment hospitalized in the oncology department of the First Affiliated Hospital of Jinan University from December 2016 to October 2017 and 18 serum sample of the healthy control.The expression level o f the VEGFR1,ALDH1A1,FAP,PDGF-AA was detected by Luminex method.By comparing the differences of marker expression level between NSCLC and control and exploring the relationship between markers and clinical pathological characteristics,including the relationship between markers,the function of markers was demonstrated that effect the invasion and metastasis in NSCLC.Results:(1)Serum ALDH1A1、FAP、PDGF-AA in NSCLC was significantly higher than that in controls(p<0.05).It was no significant differences of serum VEGFR1 between NSCLC and control(p>0.05).(2)The VEGFR1 level in NSCLC was significantly related to BMI,pleural effusion,tumor size,KPS,LY%(p<0.05);FAP serum level was significantly related to KPS,LY %(p<0.05);PDGF-AA serum level was significantly related to PLT and metastasis(p<0.05);ALDH1A1 serum level was significantly related to metastasis(p<0.05).(3)The level of VEGFR1 was negatively correlated with BMI(r=-0.328,p=0.016),KPS(r=-0.369,p=0.006)and LY%(r=-0.345,p=0.011)and positively correlated with tumor size(r=0.472,p=0.000).FAP level was negatively correlated with KPS(r=0.324,p=0.017);PDGF-AA level was positively correlated with PLT(r=0.583,p=0.000)and metastasis(r=0.356,p=0.003).ALDH1A1 was positively correlated with metastasis(r=0.468,p=0.000)and tumor size(r=0.845,p=0.000).(4)The levels of ALDH1A1 and PDGF-AA in NSCLC were positively correlated(r=0.479,p=0.000),and there was no significant correlation between VEGFR1 and FAP levels.Conclusion: NSCLC serum ALDH1A1,PDGF-AA and FAP levels were higher than healthy control,suggesting that the cytokines above may be overexpressed in NSCLC.ALDH1A1 and PDGF-AA were associated with metastasis both,and was positively related with each other,suggest that they may play an important role in the progress of invasion and metastasis.Furthermore,it is no hard to hypothesis that they may share the same or similar mechanism to promote the progress in the invasion and metastasis in lung cancer;The serum levels of VEGFR1 and FAP were not significantly correlated,suggesting that various factors may promote or inhibit the metastasis of lung cancer in the invasion and metastasis of lung cancer.The correlation between ALDH1A1 and PDGF-AA provides a new experimental evidence for the diagnosis of molecular typing of NSCLC,and provides new therapeutic targets for the treatment of NSCLC. |
