| Objective:1.To observe the behavioral effects of SPS method and SPS&S method on rats;2.To observe the effects of electroacupuncture on the learning and memory of rats with PTSD;3.To observe the effects of electroacupuncture on the expression of BDNF,GAP-43 and SYN in the hippocampus of PTSD rats for patial reveal the mechanism of electroacupuncture therapy improvethe learning and memory of PTSD rats.Methods:After adaptive breeding,50 male SD rats was randomly divided into 5 groups:control group;SPS model group;SPS with electroacupuncture group;SPS&S model group;SPS&S with electroacupuncture group.Each group had 10 rats.We used the method of SPS and SPS&S to make symptoms for the rats except the control group.EA treatment(on the points including BaiHui,ShenTing,Shen Shu)for the EA groups.Before and after the treatment we use Locomotor activity test;Morris water maze test and Elevatd plus maze test to detect the rats behavioral change.The expression of BDNF,GAP-43 and SYN protein,number of immunoreactive cells and mRNA in hippocampus were detected using Western-blotting,IHC and RT-PCR respectively.Results:1.Behavioral changes of the model groups:① There were significant differences between the Ctrl group with the SPS group(p<0.01)and the SPS&S group(p<0.01)in the Locomotor activity test;Morris water maze test and Elevatd plus maze test.②There was no statistically significant difference between the SPS group and the SPS&S group in addition to the number of crossing platforms(p>0.05),and there were significant differences in other experimental items(p<0.01).2.Behavioral changes in the treatment group:①There were significant differences in Locomotor activity test;Morris water maze test and Elevatd plus maze test between the SPS group and SPS+EA group(p<0.01),SPS&S group and SPS&S+EA(p<0.01).②There was no statistically significant difference between SPS+EA groupand SPS&S+EA group in addition to the number of crossing platforms(p>0.05),but other experimental items were significantly different(p<0.01).3.The expression changes of BDNF,GAP-43,SYN in the hippocampus of the model groups:① Ctrl with SPS group(p<0.01)and SPS&S group(p<0.01).The expression of BDNF,GAP-43 and SYN protein,number of immunoreactive cells and mRNA in hippocampus were significantly different ② SPS group with SPS&S group:There was difference in the expression of SYN in the WB test(p<0.05):in the expression of GAP-43 in the IHC test(p<0.01);significantly different in the expression of BDNF,GAP-43 and SYN mRNA(p<0.01,p<0.01,p<0.05).4.The expression changes of BDNF,GAP-43,SYN in the hippocampus of the EAgroups:①the SPS and SPS + EA group(p<0.01),with SPS&S SPS&S group +EA(p<0.01).The expression of BDNF,GAP-43 and SYN protein,number of immunoreactive cells and mRNA in hippocampus were significantly different;② There were statistically significant differences between SPS+EA group and SPS&S+EA group in the expression of BDNF,GAP-43 mRNA(p<0.01,p<0.01).5.The Escape latency was negatively correlated with BDNF,GAP-43 and SYNexpression in hippocampal.The Locomotor activity test,the percentage of Total time in target quadrant,and the Elevatd plus maze test were positively correlated with the expression of BDNF,GAP-43 and SYN expression in hippocampal.Conclusions:1.The PTSD rats made by SPS&S method had higher detection rate and more obvious bidirectional biological behavior.2.The therapy of electroacupuncture can improve the clinical symptoms of PTSD rats and improve learning and memory ability.3.The therapy of electroacupuncture can increase the expression of BDNF,GAP-43 and SYN in the hippocampus. |
