Abstract/Summary:
ObjectiveTo research the effect of Sang Xing Lung-Protection Particles(SX)on the overexpression of Th2 cells in rats with X-ray induced in pulmonary fibrosis.To clarify the mechanism of action of SX on the Radiation Pulmonary Fibrosis(RPF).Method192 male SD rats,8weeks old and average weight 200±20g were randomly divided into blank group(given normal saline 10ml/d),model group(given normal saline 10ml/d),hormone group(given prednisone tablets aqueous solution with the concentration of 0.2mg/ml,15.60ml/Kg,10ml/d),Sang Xing Lung-Protection Particles groups(given SX concentrate with the concentration of 2g/ml,5.6ml/Kg,11.25ml/Kg,22.50ml/Kg,10ml/d),except for the blank group,the other groups were given X ray chest single irradiation 14Gy,all the animals were given with gavage once a day.At designated time(12th,16th,20th,24thweek)8 rats from each group were randomly executed,acquired lung tissue.The pathological changes of lung tissue was observed by Masson staining,the Real time-Polymerase Chain Reaction was used to detected the mRNA of IL-4,IL-5 and IL-13 in lung tissue.Results1.Pulmonary fibrosis process:no fibrosis were found in lung tissue of the blank group rats;at12thh week each group rats that were irradiated still mainly showing inflammatio,at16thh week after irradiation,each group rats began to present the pulmonary fibrosis,at 24thh week end of the experiment mainly presented the pulmonary fibrosis.2.The expression of the blank group IL-4mRNA,IL-5m RNA and IL-13mRNA has been at a low level;compared with the blank group,the expression levels of each irradiation group IL-4mRNA,IL-5mRNA and IL-13mRNA increased to varying degrees(p<0.05);compared with the model group,the expression level of the hormone group and SX groups IL-4 mRNA,IL-5 mRNA and IL-13 mRNA at different points in time decreased(p<0.05);compared with the hormone group,at12th,16thh weeks,the expression of SX groups IL-4 mRNA,IL-5 mRNA and IL-13mRNA increased(p<0.05),and at20th,24thh weeks,the expression decreased;there was no difference between the SX groups(p>0.05).Conclusion1.14Gy irradiation agent could successfully establish the rats model of radiation pulmonary fibrosis,at16thh week after irradiation,each group rats all presented different degrees of pulmonary fibrosis.3.In the process of the RPF,SX can reduce the mRNA expression levels of Th2related cytokines IL-4,IL-5,IL-13 in lung tissue.4.The effect of prednisone is significantly better than the SX in early of RPF,this may be related to its strong and rapid anti-inflammatory and immunosuppressive effects.
