ObjectiveTo observe the pharmacological effect of Compound Zhenzhutiaozhi Formula(FTZ)on insulin resistance model mice induced by lipid metabolism disorder of different ages,and to verify that FTZ can improve inflammation and increase heat production by regulating the polarization of macrophages in adipose tissue.MethodThe effects of FTZ on glucose and lipid metabolism in mice of different ages were observed by using aP2-SREBP-1c transgenic mice of different ages.The plasma glucose in mice before and after administration was detected by routine biochemical method.The content of triglyceride,total cholesterol and low density lipoprotein cholesterol.The changes of glucose tolerance and insulin resistance in mice of different age groups were observed by oral glucose tolerance test(OGTT).The content of fatty acids in the liver was measured by GC-MS,the polarization of macrophages in adipose tissue was detected by flow cytometry,and the mrna expression level of target gene in adipose tissue was detected by reverse transcription-polymerase chain reaction(RT-PCR).The aP2-SREBP-1c mice was mainly manifested as white fat atrophy,disorder of glucose and lipid metabolism,obvious insulin resistance and fatty liver,and the increase of age aggravated the damage of glycolipid metabolism.FTZ can improve blood glucose,total cholesterol,insulin level and glucose tolerance of mice in different age groups.Pathological examination and lipid content measurement showed that FTZ could reduce lipid ectopic deposition and improve fatty liver.GC-MS test showed that FTZ could regulate fatty acid metabolism.The results of flow cytometry showed that FTZ could regulate the level of M1 and M2 macrophages and improve the inflammatory infiltration of adipose tissue.RT-PCR results showed that TNF-? and il-6 were significantly down-regulated in subcutaneous adipose tissue.The expression of inflammatory inhibitors such as IL-10,IL-4 and IL-13 were significantly increased.ConclusionFTZ can significantly regulate the level of glucose and lipid metabolism,decrease the ectopic deposition of lipids in liver,decrease the level of fatty acids and glycosylation in the liver of insulin resistant model mice induced by adipose metabolic disorders of different ages.It can regulate adipose tissue macrophage polarization,increase heat production,balance body energy metabolism,increase insulin sensitivity and improve insulin resistance.
