| Callicarpa nudiflora Hook.et Arn.belongs to Verbenaceae Callicarpa.The medicinal parts of this plant are leaves and twigs.Though this plant had been used as medicine for a long period,the quality of C.nudiflora materials is not stable enough for industrial uses.Furthermore,quality control method for this plant has not been developed yet.Based on this,we selected the main anti-inflammation and hemostatic chemical constituents in C.nudiflora for analysis.We established methods using HPLC and UV to determinate the contents of these chemical compounds and the extraction method of C.nudiflora was optimized.Then we compared some chemical composition contains of the samples obtained from different places of origin,harvesting periods and post-harvest processing methods.The results in present study may provide basis for enhancing production and processing this herb.The main conclusions are as follows:1.The method for determining the contents of seven monomer components and three effective parts in C.nudiflora was established,and the method for extracting the main components of C.nudiflora was optimized.A method for the determination of 7 monomer components(caffeic acid,forsythiaside B,luteolin,verbascoside,islataside,apigenin-7-O-β-D glucoside,luteolin)and 3 effective fractions(total flavonoids,total phenolic acids,total saponins)of C.nudiflora.was established by HPLC and UV respectively.The established method for the determination of HPLC content has a good separation degree and good linearity(R~2≥0.9990).The average recovery rate is between 96.4% and 105.5%.The established UV detection method has good linearity for each effective part(R~2≥0.9932).The recovery rate of the sample was between 98.88% and 104.12%.Response surface method was used to optimize the extraction method of C.nudiflora based on the comprehensive scores of three effective sites and extract yields.The optimal extraction time was42 min,the ratio of solid to liquid was 1:17,and the ethanol concentration was 62%,extracted 2 times.2.There are significant differences in the contents of major chemical constituents of C.nudiflora from different habitats and climatic regionsThe variance analysis and principal component analysis were used to analyze the content of 7 monomer components in C.nudiflora samples from 93 different producing areas in Hainan,Guangdong and Guangxi.It was found that the total content of 7 monomer components of C.nudiflora in Hainan was higher than that of the samples outside the province(P>0.05),and the average content of 7 monomer components was 25.120 mg/g in Hainan and 12.640 mg/g in outside the province;9 component samples in Hainan can be clearly divided into two types:the IATa(Ding’an,Tunchang,Haikou,Lingao)in the northern marginal tropical humid area and the IATb(Baisha,Qiongzhong,Wuzhishan,Dongfang,Ledong)in the central humid mountainous area.The content of each component was higher in the IATb sample(P>0.05).The IATb area was 30.694 mg/g and the IATa area was 15.212 mg/g on the average content of the seven monomer components.3.There are significant differences in the contents of major chemical constituents of C.nudiflora in different harvesting parts and harvesting periodsTo determinate content of major constituents of C.nudiflora in three different parts of functional leaves,young leaves and young shoots and functional leaves ten of harvested dates,Analysis of variance showed that the total content of the three effective parts,the total content of the seven monomer components,and the yield of extracts were all in the functional leaves>young leaves>shoots(P<0.05),and the functional leaf were 216.725 mg/g,38.531 mg/g,31.4%.The total content of three effective parts,the total content of seven monomer components and the extraction yield of functional leaves in the 10 harvest periods were the highest in the early fruit set,which were 294.533 mg/g,72.240 mg/g and 42.1%respectively.The content is the lowest in the late fruit drop stage,and the content of its main chemical components first increases and then decreases.4.Different primary processing methods have a significant effect on the content of major chemical component of C.nudifloraThe samples of C.nudiflora were dry in the shade,dried at different temperatures and dried at different temperatures after green removing 5 min,the main chemical compositions of C.nudiflora were characterized by variance analysis.It was found that the main chemical composition content of the samples dried after green removing 5 min was generally higher than direct drying and dried in shade samples.The drying processes of 55℃ of after green removing 5 min,the main chemical component content is highest for the best temperature which total content of the three effect sites,the total content of the seven monomer components and the extract yield were 574.254 mg/g,103.085 mg/g and42.7% respectively. |
