Objective: To observe the regulating effect of warming kidney yang decoction on monoamine neurotransmitters in CUMS rat model of depression and the effects on the expression of CREB and BDNF receptor signal transduction mechanism in the post,and explore its mechanism of action.Methods: 60 healthy SPF male SD rats were randomly divided into 4 groups,each group was divided into 15 groups: normal control group,depression model group,experimental group and fluoxertine hydrochloride group.The model of depression induced by chronic unpredictable mild stress(CUMS).After the model of rats in the open field test the success of modeling.After 21 days of intragastric administration,the normal control group and the depression model group were given normal saline.After the administration,the open field test evaluation of depression model rats behavioral changes,Hippocampal Monoamine Neurotransmitter of rats was detected by ELISA(5-HT,DA,NE),using RT-PCR method to detect the expression of CREB and BDNF in hippocampus of rats.Results:1.The experimental results of open field test: except the normal control group,the rats after 21 days of chronic mild chronic unpredictable mild stress(CUMS)after stimulation showed a different degree of depression like changes.The open field test in the model group after modeling level and vertical score scores were significantly lower(P<0.01),suggesting that CUMS induced decline in exercise capacity in rats and explore the ability of modeling.The rats after gastric filling treatment after 21 days,the depressive behaviors of Warming kidney yang group and fluoxetine group rats were significantly improved,the vertical score of Warming kidney yang group and fluoxetine group rats score,field experiment was significantly improved,compared with the depression model group,with significant difference(P<0.01).2.ELISA method for detection of Rat Hippocampal Monoamine Neurotransmitters in the depression model rats hippocampal 5-HT,DA and NE decreased significantly compared with normal control group,there was significant difference(P<0.01).Warming kidney yang group and depression model group compared to Warming kidney yang can significantly improve 5-HT,DA and NE content in hippocampus of rat model of depression,the difference was statistically significant(P<0.05),but the hippocampus Warming kidney yang group rats in DA and NE were compared with normal control group,with significant difference(P<0.05),depression model group,Warming kidney yang group and fluoxetine group rats hippocampus 5-HIAA content decreased while visible,but compared with the normal control group,no significant difference.3.RT-PCR method was used to detect the expression of CREB and BDNF: the expression of CREB and BDNF in hippocampus of depression model group was significantly lower than that of normal control group(m RNA)(P<0.01).Warming kidney yang group rats hippocampus CREB,BDNF expression of m RNA increased significantly compared with the depression model group,with significant difference(P<0.05,P<0.01),Warming kidney yang group BDNF m RNA expression compared with normal control group,with significant difference(P<0.05),illustrated by giving21 days Warming kidney yang treatment the expression of CREB and BDNF in hippocampus m RNA upregulation of rat model of depression,improve the depression in rats,but the expression of m RNA BDNF did not recover to the level of health.The drug group was compared among depression model rat hippocampal CREB and BDNF expression of m RNA had no significant difference,indicating Warming kidney yang and fluoxetine hydrochloride in improving BDNF m RNA and CREB in hippocampus of rat model of depression effect to expression.Conclusion:the anti depression effect of warming kidney yang prescription is clear.CUMS can significantly improve the rat model of depression.Open field test scores,can improve the CUMS in hippocampus of depression model rats 5-HT,DA,NE and BDNF were up-regulated CREB expression level of m RNA,improve the depression in rats.
