Objective:Study on the mechanism and clinical effects of Buyang Huanwu Decoction on pulmonary fibrosis induced by bleomycin in rats with an important pathway protein mTOR.Method:Methods 144 male SD rats weighing more than 200 20 g were randomly divided into control group,model group,prednisone group,Buyang Huanwu Decoction(high,middle and low dose group),with each group of 24 rats in each group.The blank group did not do any treatment,the rest of the group will be injected bleomycin method to build a pathological model of pulmonary fibrosis.After modeling 24 h,given the corresponding medicine in each group were killed by exsanguination at seventh,28 days,8 rats were randomly selected for lung tissue sampling,using HE and Masson were observed in lung tissue and alveolar inflammation,fibrosis staining,immunohistochemical stain group into pathology(chain mycin avidin peroxidase.SABC)distribution sections of lung mTOR protein expressiResult:General situation:The rats in the blank group were normal in body shape,and were more sensitive to external stimuli.The rats in the model group showed lassitude,dull eyes,quick decline,shortness of breath,some mice even less tired lying motionless,coarse hair withered,appetite decreased,emaciation.The rats in the other groups were better in spirit,sensitive in reaction,normal in breath rhythm and frequency.The activity ability and the amount of food intake decreased with time.Pathomorphology:HE staining: normal control group lung tissue of rats had no obvious lesion morphology;compared with the control group,the alveolar structure of rats in the model group was destroyed,found a large number of inflammatory cells and the proliferation of collagen fibers;compared with the model group at different time points in each group the alveolar inflammation and the degree of pulmonary fibrosis were improved to a certain extent.In the Masson staining group,the collagen layer in the bronchial wall was thinner,and only a few blue collagen fibers were found in the alveolar area.The 7d model group rats appeared obvious alveolar thickening in the bronchial wall,alveolar septa were found to have blue collagen fibers;the 28 d in the bronchioles and fibrosis area of consolidation found a large number of blue collagen fibers,there is diffuse pulmonary fibrosis.Prednisone group and Buyang HuanwuMTOR: compared with blank group,the content of TNF-(BALF)in bronchoalveolar lavage fluid(P)of model group was increased at different time points(< 0.05).The content of mTOR decreased at day 28,compared with 7 days with significant difference(P < 0.05);compared with the model group,prednisone group at each time point and the positive also five soup high,medium and low dose group were decreased in rats in the bronchoalveolar lavage fluid(BALF)in the content of mTOR(P < 0.05);comparison with the prednisone group,the seventh day Yang also five soup in each dose group had significant difference(P < 0.05);comparison between the five Buyanghuanwu decoction each dose group,high dose group at each time point and the difference between the low and medium dose group(P < 0.05) empirical conclusion:Abnormal expression of mTOR in the course of pulmonary fibrosis.Buyang Huanwu Decoction five can effectively inhibit the excessive activation of mTOR,thus play a role in the intervention of pulmonary fibrosis.
