| Tibetan medicine as the world recognized traditional high altitude medicine,has a complete and scientific system of theory and clinical practice,which derived from the production and living practice of Tibetan people.It is the confluence of Tibetan ancient and modern medical experience.The introduction and localization of exotic medicine under the background of Silk Road,make the Tibetan medicine system more abundant and perfect.It absorbed the essence of ancient Chinese medicine,Ayurvedic medicine and Arabia medicine and so on in the long course of development,formed a diverse medical culture,theoretical foundation and clinical technology.San-Guo Tang San is undoubtedly the most commonly used and diversified drug in Tibetan medicine,named“Zhe Bo Song Tang”,with the effects of clearing heat and regulating the Long and blood for the treatment of plague fever and overwork.In the Tibetan medicine clinical San-Guo Tang San should taken before blood-letting therapy 3-5 days to separate healthy blood and bad blood,it can significantly enhance the effect of blood-letting therapy.Clinical studies suggest the impressive effect of San-Guo Tang San with blood-letting therapy on the treatment of high altitude polycythemia(HAPC),but it is still lack of systematic scientific research from the view of molecular mechanism about HAPC.The primary cause of HAPC is hypoxia,and the express of HIF-1α,EPO,ROS are closely associate with hypoxia,and there is a correlation between them.Therefore,whether HIF-1α/EPO/ROS is Anti-HAPC signaling pathways of San-Guo Tang San,And how to return modern microcosm to the macro-Tibetan basic theory,It is an important part of this paper.Objective:1.untangle the history and cultural connotation of San-Guo Tang San,to analyze its medication rules of Tibetan medicine,to provide theoretical basis of Tibetan Medicine for the study of its mechanism.2.To investigate the anti-hypoxia effect of San Guo Tang San and its pharmacological effects on HAPC model rats,to research the mechanism of HAPC treatment.Methods:1.Literature mining:using the method of literature analysis and key personnel interviews,to gathering the information about the application and dissemination of San-Guo Tang San in the countries along the Silk Road,and analysis combined with historical background.2.Data mining:TCMISS(V2.5)was used to build a prescription database of Tibetan medicine containing chebula or“San-Guo”,use the software statistical statement module,association rules and improved mutual information method to analyze the law of medication.3.Pharmacological experimental:(1)Using normal mice to conduct experimental study on the hypoxia tolerance,the survival time of mice in each group was recorded to observe the anti-hypoxia effect of San-Guo Tang San.(2)Using the Hypoxia chamber to establish the rat model of HAPC,testing the indexes that include blood routine,blood rheology and SOD,MDA,EPO,ROS,HIF-1α,et al.after medication,To explore the therapeutic effect and potential mechanism of Tibetan San-Guo Tang San on HAPC model rats.Results:1.Literature mining results:San-Guo Tang San is a typical representative of Multiracial cross drug from ancient India.Through the ancient Silk Road spread to a number of nations.In India,Arabia,China’s traditional medicine and ethnic medicine are all used,accompanied by different names and Applications.2.Data mining result: The Clinical application of San-Guo is extremely broad,whose ingredients mostly had the functions of clearing heat and detoxicating,promoting blood circulation and stopping pain,warming the middle-jiao and promoting the circulation of Qi.And the main treatment of diseases is “Tripa”disease,heat syndrome,blood heat,gout,eyes disease,white vein disease.3.Pharmacological experimental results:San-Guo Tang San can prolong the survival time of mice at normal pressure and hypoxia.The blood index detection of HAPC model rats is displayed that RBC,HGB,HCT and whole blood viscosity were increased in different degrees,and the blood indexes of the drug group decreased in different degrees.Detection of hypoxia related cytokines showed that San-Guo Tang can make EPO,SOD,MDA,ROS,HIF-1α,EPO mRNA and other indicators of HAPC model rats have improved.Conclusions:1.Tibetan medicine San-Guo Tang San has a long history and rich cultural connotation,It can be used as an example of the national culture and medicine exchanges along the Silk Road to promote the further cooperation of traditional medicine.2.According to Tibetan medicine theory,using data mining techniques to analyze the medication rules of San-Guo Tang San,found that it is more compatible with drug can clearing heat and detoxicating,promoting blood circulation.It is consistent with treatment requirements of HAPC.3.San-Guo Tang San has a good anti-hypoxia effect,can reduce the rats’ red blood cell count and blood viscosity,and it can prevent HAPC due to high altitude hypoxia,the mechanism may be related to the "ROS-HIF-1α-EPO pathway". |
