Objective:To investigate the effect of Transforming growth factorβ1(TGF-β)on the expression of autophagy related protein Beclin1 in cervical cancer SiHa cells.Methods:Six-well plates were inoculated with cervical cancer SiHa cells at a logarithmic growth phase at a concentration of 1.5 x 105/ml,adhered to the cells after 24 hours of routine culture,and starved overnight.SiHa cell has been divided into the following groups,TGF-β1 concentration groups: 0,5,10ng/m L,incubating for 24 hours;TGF-β1 10ng/m L action time group:0h,12 h,24h.Real-time fluorescence quantitative PCR and Western blotting were used to detect the Beclin-1 mRNA and protein expression levels.Results:1.After 24 hours of different concentrations of TGF-β1 on cervical cancer cells,the expression of Beclin1 protein and mRNA increased significantly with the increase of TGF-β1 concentration(P<0.05).2.10ng/ml TGF-β1 acted on cervical cancer cells,and the expression of Beclin1 protein and mRNA increased significantly with the prolongation of time(P<0.05).Conclusion:Exogenous TGF-β1 can promote expression of Beclin1 mRNA and protein in SiHa cells of cervical cancer.With the certain density and action time,TGF-β1 can promote the expressions of Beclin1 and there are density-effect and time-effect relationship.
