Objective 1.The population-based family study method was used to study the distribution characteristics of environmental risk factors in the case family and the control family group,the first-degree relatives of the case family and the first-degree relatives of the control family,and the impact of environmental factors on the occurrence of tuberculosis was analyzed.2.Applying molecular epidemiological methods to investigate the genetic effects of the genetic variants of IL-10 gene rs1800629,IFN-? gene rs2430561 and TNF-? gene rs1800896,and analyze the impact of the interaction between genes and genes and genes and the environment on tuberculosis.MethodsFrom the perspective of cytokines,this study investigated the association between gene polymorphisms of the IL-10 gene rs1800629,FN-? gene rs2430561 and TNF-? gene rs1800896 and the susceptibility to tuberculosis by the population-based family study.the frequency of genotypes and alleles for each studied gene was calculated,and the distribution differences between the case family and control family,including their first-degree relatives,were analyzed.The impact of different genes on tuberculosis incidence was analyzed.Logistic regression model was used to analyze the association among environmental factors,genetic variation of the studied genes and the risk of tuberculosis.Family clustering in the case family population and comparison of the epidemiological characteristics of each group were analysed.Multiplication interaction method based on Logistic regression model was used to calculate the index of interaction between genes and genes and genes and the environment.The bootstrapping re-sampling program was used to calculate additive interaction indices of genes-genes and genes-evironment,and the environmental risk factors and genetic factors in the occurrence of tuberculosis were evaluate comprehensively.ResultsA total of 180 samples were collected in this study,of which 89 people were in the case family,and the number of case families were 28.The number of patients was 41 and the first-degree relatives were 23.The control family group had a total of 91 people,which came from 35 families.The distribution of BMI index,education level,place of residence and fitness activity in the case-families group and the control family group were different.The ratio of BMI was <18.5,the education level was high school,secondary school and below,and the place of residence in rural areas and no fitness activities in the case family was higher than the control family group.The distribution of BMI index and fitness activities in the first-degree relatives of the case-families group and the control-family group were different.The ratio of BMI < 18.5 and non-fitness activities of he first-degree relatives of the case-families group was higher than the control-family group.The distribution of smoking and alcohol consumption in the case family was different between the non-case group and the case group.Smoking and drinking increased the risk of tuberculosis(OR=2.89,95%CI=1.07-7.76;OR=2.89,95%CI=1.07-7.76).).There were differences in distribution of fitness activities between the cases of first-degree relatives and non-cases in the case family.Fitness activities could reduce the risk of tuberculosis in first-degree relatives(no fitness activity vs fitness activity: OR=4.29,95% CI=1.04-7.80).The rs1800896 of the TNF-? gene may be a susceptibility factor for tuberculosis.Compared with the A allele of TNF-? gene rs1800896 carrier,the risk of tuberculosis in individuals carrying the G allele is significantly higher.Conclusion 1.The factors of BMI < 18.5,high school education,technical secondary school and below,living in rural areas,smoking,drinking,and non-fitness activities may be influence to the development of tuberculosis.2.The rs1800896 of the TNF-? gene may be a susceptible locus for tuberculosis.Compared with the A allele of TNF-? gene rs1800896 carriers,the risk of tuberculosis in individuals carrying the G allele is significantly higher.The results of this study were obtained from a small sample of epidemiological studies.Subsequent increases in sample size are still needed to further verify.
