Objective:Diarrhea in children is caused by multiple factors and multi-pathogen,which is based on a group of diarrhea-predominant symptoms,age of onset is 2 years old,less than 1 year of age accounted for about 50%.Children around the world die from diarrhea every day up to 5 million to 18 million.In China,second only to respiratory infections in children with diarrhea are the second common disease.DingGuier umbilicus patch are Cloves,Cinnamon,Piper longum as the main ingredient,combined with lanolin,azone,white petrolatum,glycerin,paraffin and other accessories,an auxiliary treatment of infantile diarrhea in children commonly used drugs.From the perspective of traditional Chinese medicine prescription studies,Cloves,Cinnamon,Piper longum belong to The Interior Warming Chinese Medicinal Herbs.Moreover,Cloves,Cinnamon and Piper longum have the ability to warm up the interior and dispel cold to act on internal conditions,can be used to treat the deficiency and coldness of the spleen and stomach marked by cold pain in epigastric abdomen.On effective constituents and pharmacological study of each single herb Cloves,Cinnamon,Piper longum,the treatment of infantile diarrheaby DingGuier umbilicus patch are common,but the study on the main active components and the prescription of DingGuier umbilicus patch are little.On the basis of previous reports in the literature,by made the the supercritical fluid extraction of Cloves,Cinnamon,Piper longum and the mixture for umbilicus patch,to determine the main active ingredient in DingGuier umbilicus patch.Furthermore,the main active component of GC/MS analysis method is established,and the scientific DingGuier umbilicus patch is explored by transdermal absorption experiment.The purpose of this study is to confirm the main active component of DingGuier umbilicus patch and establish the GC/MS analysis method of the main active component,and lay the foundation of scientific prescription on DingGuier umbilicus patch.Methods:Cloves,Cinnamon,Piper longum and DingGuier umbilical paste prescriptions by CO₂supercritical fluid extraction respectively,been extracted product,which was using GC chromatography,and combined with the full scan Scan Qualitative analysis by GC/MS,to determine the group of the main active ingredient.The patch were made from the extracted product,by in vitro percutaneous absorption experiments,using GC/MS analysis of transdermal ingredients in released solution.The results of Cloves supercritical extraction product by GC/MS Scan:Eugenol was detected at 8.506 min,the peak area was 66.82%;Caryophyllene was detected at 9.604 min,the peak area was 8.52%;2,4-di-tert-butyl-phenol was detected at 11.183 min,the peak area was 1.98%;Eugenol acetate ester was detected at 11.533 min,the peak area was22.68%.The results of Cinnamon supercritical extraction product by GC/MS Scan:Cinnamic aldehyde?E?was detected at 7.493 min,the peak area was 53.90%;Eugenol was detected at 8.534 min,the peak area was 19.28%;Caryophyllene was detected at 9.610 min,the peak area was 6.88%;Eugenol acetate ester was detected at 11.550 min,the peak area was6.17%;2H-1-Benzopyran-2-one was detected at 9.907 min,the peak area was 4.43%;2-Propenal,3-?2-methoxyphenyl?was detected at 11.681 min,the peak area was 1.48%;Copaene was detected at 8.860 min,the peak area was 1.34%;alpha-Caryophyllene was detected at10.216 min,the peak area was 1.01;.alpha.-Muurolene was detected at11.069 min,the peak area was 1.04%.Results:The results of Piper longum supercritical extraction product by GC/MS Scan:Cinnamic aldehyde?E?was detected at 7.430 min,the peak area was 26.82%;2,4-di-tert-butyl-phenol was detected at 11.184 min,the peak area was 22.61%;piperine was detected at 31.806 min,the peak area was 10.13%;Caryophyllene was detected at 9.604min,the peak area was 2.01%;Caryophyllene was detected at 9.604 min,the peak area was2.01%;Eugenol acetate ester was detected at 11.533 min,the peak area was 1.17%;Hexadecanoic acid,methyl ester was detected at 23.022 min,the peak area was 1.96;Octadecanoic acid,methyl ester was detected at 25.036 min,the peak area was 2.03%.The results of the mixture supercritical extraction product by GC/MS Scan:Eugenol was detected at 8.517 min,the peak area was 65.29%;Phenol,2-methoxy-4-?2-propenyl?-,acetate was detected at 11.538 min,the peak area was 11.99%;Cinnamic aldehyde was detected at 7.424 min,the peak area was 7.21%;Piperine was detected at 31.811 min,the peak area was 2.84%;Caryo-phyllene was detected at 9.604 min,the peak area was 5.36%;2,4-di-tert-butylphenol was detected at 11.184 min,the peak area was 1.03%;Germacrene was detected at 10.737 min,the peak area was 1.18%;in addition,.alpha-Caryophyllene,Isocaryophillene and Copaene were detected.The results of percutaneous absorption in vitro:the main active ingredients of Cloves transdermal patch were Eugenol,Phenol,2-methoxy-4-?2-propenyl?-,acetate;the main active ingredients of Cinnamic transdermal patch were Cinnamic aldehyde,2-Propenal,3-?2-methoxyphenyl?-;there were not the main active ingredients in Piper longum transdermal patch;the main active ingredients of DingGuier umbilicus patch were Eugenol,Phenol,2-methoxy-4-?2-propenyl?-acetate,Cinnamic aldehyde,2-Propenal,3-?2-methoxy-phenyl?-Benzenepropanoic acid,and Piperine.Conclusion:The main active ingredients of DingGuier umbilicus patch include Eugenol,Phenol,2-methoxy-4-?2-propenyl?-,acetate,Cinnamic aldehyde,2-Propenal,3-?2-methoxy phenyl?Benzenepropanoic acid,and Piperine.The results on percutaneous absorption in vitro experiment can be seen,DingGuier umbilicus patch contain the main active ingredient in all of the above.Only after the composition of Piper longum,cinnamon and cloves prescription,can make the active ingredient Piperine transdermaled.The experimental results of in vitro percutaneous release search obtained DingGuier umbilicus patch prescriptions were scientificity.
