Expression And Clinical Significance Of Caspase-8 And Caspase-9 In Gastric Carcinoma

Gastric carcinoma(GC)is one of the most commmom malignant tumors in the world,and the incidence of east Asia is the highest in the world.Recent years,due to the development of our country's economy,the improvement of people's living standards,the change of the diet structure and the environment,the aging of population,GC incidence and mortality rates are the highest in the world.Gastric cancer occult in onset,many GC patients belong to advanced stage when they are diagnosed,so their prognosis are poor.The key of improving the prognosis of GC is improving the early diagnostic rate.GC is the result of multiple genes and multiple factors,so its pathogenesis is still not clear.At present,many literatures have reported that the imbalance of cell proliferation and apoptosis is related to the occurrence of tumor,apoptosis plays an important role in the cell death.So the mechanism of apoptosis has become a hot research topic.Caspase protease family that exists in the cytoplasm is vital to the regulation of cell apoptosis.Caspase that exists in the inactive zymogen is widely distributed in human normal tissues,for example liver?colorectal?thyroid?stomach?muscle,and so on.Caspase-9 and Caspase-8 belong to the apoptotic promoter that can be activated or activated by other proteins in the activation and activation of the downstream Caspase,which lead to cell apoptosis.Recent years,Caspase-8 and Caspase-9 were found to show low expression in many tumor tissues.They are related to the occurrence,development and prognosis of tumor,such as thyroid cancer,colorectal cancer,gastric cancer,pancreatic cancer,and so on.At present,there are few reports about the short-term prognosis of gastric cancer with Caspase-8 and Caspase-9 at home and abroad.This experiment,which was based on gastric cancer tissue and pericarcinous tissue samples of patients with gastric carcinoma and detected the expressions of Caspase-8,Caspase-9 protein,analyzed the relationship between expressions of Caspase-8,Caspase-9 protein and the development and clinicopathological feature of gastric carcinoma,and explored if there was relativity between the expressions of Caspase-8,Caspase-9 protein and the short-term prognosis of gastric carcinoma.objective: The experiment test the expression of Caspase-8 and Caspase-9 protein in gastric carcinoma tissues.Our aim is to investigate the association of Caspase-8 and Caspase-9 protein with Clinicopathological features,and to explore the relationship between the expression of protein and the short-term prognosis of gastric cancer.Methods: This experiment utilize immunohistochemistry to detect the expression of Caspase-8 and Caspase-9 protein in gastric cancer tissues and adjacent tissues of gastric cancer.Finally,clinical and pathological data of gastric cancer patients were collected,and the date were analyzed.Results: The expression of Caspase-8 and Caspase-9 protein are positive in gastric cancer and adjacent tissues.The positive rate of Caspase-8 protein in gastric cancer is 32.43% and in adjacent tissues is 72.97%.The positive rate of Caspase-8 protein in gastric cancer tissue is significantly lower than that in the adjacent tissues(P<0.05).The positive rate of Caspase-9 protein in gastric cancer is 37.84% and in adjacent tissues is 62.16%.The positive rate of Caspase-9 protein in gastric cancer tissue is significantly lower than that in the adjacent tissues(P<0.05).In 37 cases,the expression of Caspase-8 protein is correlated to degree of differentiation?lymph-node metastases and survive time(P<0.05),there is no correlation with patients' age,sex,tumor size,tumor location,the depth of tumor invasion,TNM stage(P>0.05);the expression of Caspase-9 protein correlated with degree of tumor differentiation?lymph-node metastases?the depth of tumor invasion?TNM stage?survive time(P<0.05),but not correlated with age?sex?tumor size?tumor location(P>0.05).Conclusion: Caspase-8 and Caspase-9 are low expressed in gastric cancer,and participating in the occurrence and development of gastric cancer.Caspase-8 and Caspase-9 are related to the short-term prognosis of gastric cancer.
