Objective:1.To investigate the effect of Mannan-binding lectin(MBL)on proliferation and secretion of TNF-β、IL-6、IL-8、IL-17 and the expression of MBL and MAS P2 of mesangial cells culturing under high glucose condition.2.Observing the effects of triptergium wilfordii on the expression of MBL,MAS P2 and inflammatory cytokine of mesangial cells culturing under high glucose condition to explore the potential protective mechanism of triptergium wilfordii on diabetic nephropathy.Method:20 male SD rats were randomly divided into TW group(10)and normal contro l group(10)after 2 weeks adaptative diet,the TW group were treated with tript ergium wilfordii(10mg·kg-1·d-1)for 1 week;After culturing the mesangial cells i n high glucose condition,we tested the effects of MBL on proliferation of cell s by CCK-8.And then the secretion of related inflammatory factors were meas ured by the enzyme linked;Real-time PCR was used to detect the expression of MBL and MASP2 mRNA of mesangial cells while Western blot was used to detect the expression of MBL and MASP2 protein.Result:1.Compared with Con group,the proliferation of mesangial cells was significantl y inscreased in high glucose condition,and MBL enhanced the growth of the mesangial cells.2.ELISA results suggest:compared with Con group,high glucose prompted the se cretion of IL-6,IL-8,IL-17 and TNF-β,and MBL aggravated these inflammato ry factors releasing.3.Real-time PCR results suggest:Expression of MBL and MASP2 mRNA of me sangial cells in high glucose condition were significantly inscreased while comp ared with Con group;Expression of MBL and MASP2 mRNA of mesangial cell s in TW group was lower compared with cells in high glucose condition.Weste rn blot results suggest:Expression of MBL and MASP2 protein of mesangial ce lls in high glucose condition were significantly inscreased while compared with Con group;Expression of MBL and MASP2 protein of mesangial cells in TW group was lower compared with cells in high glucose condition.Conclusion:1.The proliferation of mesangial cells was significantly inscreased in high gluco se condition,and MBL enhanced the growth of the mesangial cells2.The Mannan-binding lectin pathway take part in the inflammatory reaction of mesangial cells in high glucose condition.3.Triptergium wilfordii may protect the mesangial cells by inhibiting the Manna n-binding lectin pathway to reduce the expression of inflammatory factors.
