| Objective To observe the effects of resveratrol on body-weight of high-fat diet fed rats,and investigate the effe ets on the expression of FoxO1 and UCP1 in adipose tissue,in order to explore new ways of thinking for the prevention and treatment of obesity.Methods Healthy three-week-old Sprague-Dawley(SD)rats(48)were randomly divided into 4 groups,each of groups consisted of 12,①Standard diet controlled group(group SC),②High-fat diet controlled group(group HFC),③High-fat diet + 6mg/kg/d resveratrol group(group Res1),④High-fat diet + 18mg/kg/d resveratrol group(group Res2).Feds with standard diet to group SC,and the other groups fed with high-fat diet.At the same time of obesity models establishing,drug intervention started.Group SC and group HFC were absorbed with saline water,Group Resl and group Res2 were administered taken with resveratrol.To measure weight and size of rats every week,which is able to calculate the Lee’s index.All rats were sacrificed at the end of 11th week after drug intervention.Serum samples were collected before sacrifice to determine the levels of bone morphogenetic protein 7(BMP7)by ELISA;took white adipose tissue around genitals to weighed and made HE slice of pathology to observe the morphological changes of white adipose tissue;the expression of silent mating type information regulation 2 homolog 1(Sirt1),Forkhead box O-1(FoxO1)and Ser-ine/threonine kinase(Akt)were detected by ELISA and RT-PCR in white adipose tissue around genitals;and the expression of uncoupling protein-1(UCP1)and BMP7 were detected by ELISA and RT-PCR in interscapular brown adipose tissue(BAT)and subcutaneous white adipose tissue(WAT).Results 1.There were no significant difference of body-weight and the Lee’s index between four groups before obesity models establishing(p>0.05);after fed with high-fat diet,body-weight and Lee’s index were increased extremely in group HFC,compared with group SC(p<0.01),compared with group HFC,body-weight and Lee’s index were decreased significantly in group Res1 and Res2(p<0.01),and the mean values were not diffient from group SC;compared with the same group at the beginning of models establishing,the Lee’s index is significantly increased in HFC group since 8th week after fed with high-fat diet(P<0.05,P<0.01),while the Lee’s index in SC,Resl,Res2 group shows no differences in any stage(P>0.05).2.Compared with group SC,the weight of white adipose tissue around genitals and interscapular brown adipose tissue were increased dramatically in group HFC(p<0.01);compared with group HFC,the weight of white adipose tissue around genitals and interscapular brown adipose tissue were decreased significantly in Group Resl and Res2(p<0.01,P<0.05);3.Compared with group SC,the levels of Sirtl mRNA in WAT were decreased significantly in Group HFC(p<0.01);compared with group HFC,the levels of Sirtl mRNA were increased dramatically in group Res2(p<0.01),but it was always lower than it in group SC(p<0.01);4.The levels of Akt mRNA in WAT were increased extremely in group HFC,compared with group SC(p<0.01);but compared with group HFC,the levels of Akt mRNA were decreased significantly in Group Resl and Res2(p<0.05,p<0.01);5.Compared with group SC,the levels of FoxO1 mRNA in WAT were decreased dramatically in group HFC(p<0.01);compared with group HFC,the levels of FoxO1 mRNA were increased extremely in Group Resl and Res2(p<0.05,p<0.01),and the mean values were not diffient from group SC;6.The levels of UCP1 mRNA in WAT and BAT were decreased significantly in group HFC,compared with group SC(p<0.05,p<0.01);but compared with group HFC,levels of UCP1 mRNA in WAT and BAT were increased dramatically in Group Res1 and Res2(p<0.01,p<0.05),and the mean values were not diffient from group SC;7.The levels of BMP7 in serum were decreased extremely in group HFC,compared with group SC(p<0.05);but compared with group HFC,the levels of BMP7 in serum were increased significantly in Group Res2(p<0.01),and it was higher than it in group SC(p<0.01);8.The expression of UCP1 in tissue homogenate of WAT were positively related to the expression of BMP7 in serum and in tissue homogenate of WAT(r=0.473,p=0.001;r=0.302,p=0.037);the expression of UCP1 in tissue homogenate of BAT were positively related to the expression of BMP7 in serum(r=0.563,p-0.000).Conclusion 1.High-fat diet fedding has effects on suppression of Sirtl’s secretion and the increasing expression of Akt in rats’ WAT,however to elicit down-regulation the expression of FoxO1;2.Resveratrol treatment could reduce body-weight of high-fat diet induced rats by increasing the expression of FoxO1,which is not only correlated with inducing deacetylation of Sirtl,but also may corporately via inhibiting PI3K-Akt phosphorylation signal pathway;3.Resveratrol treatment could increasing the expression of UCP1 in WAT via promoting the expression of BMP7 in serum and WAT. |
