Purpose:In this study,we Based on the NAFLD animal model,observe the impacts of pomegranate polyphenols to liver function,discuss the efficiency of pomegranate to the hepatic function and blood fat,furnish the experimental documents for study of effects mechanism by pomegranate polyphenols.And,using high performance liquid chromatography(HPLC)determination the gallic acid and ellagic acid content in pomegranate polyphenols,to provide quality control,and laboratory data for the determination of the chemical composition of the main ingredients of pomegranate.Methods:at 20 ApoE-/-mice and 10 C57B6/L mouse were randomly divided to control group,modeling group and PFP group,afterl week adaptive feeding in the SPF level animal experiment house the animals in the modeling group and PFP group were fed with high fat diets for 8 weeks to built NAFLD animal model,after that we orally administrated pomegranate polyphenols(140mg/kg)to the PFP group animals as well as same volume of distilled water to the animals in control grroup and modeling group.after 8 week PFP,we test fasting blood glucose,collect blood samples of all animals,detect insulin level of blood samples,blood lipid and aminotransferase levels as well as pathological changes of liver pathology.then determination of total polyphenol content in pomegranate polyphenols by UV spectrophotometry,Then using Waters Symmetry Shield RP18 column(250mmx4.6mm×5um)use methanol-0.05 phosphoric acid solution(5:95)as the mobile phase,velocity of flow 1.0ml/min,the detection wavelength 273nm,column temperature 25℃,the injection volume 1Oul,calculation of gallic acid peak area,using Waters Symmetry Shield RP18 column(250mm×4.6mm×5um),Methanol aqueous-0.3 phosphoric acid(55:45)as the mobile phase,velocity of flow 1.Oml/min,the detection wavelength 254nm,Column temperature 35℃.the injection volume lOul,calculation of gallic acid peak area.Result:compared with model group,the blood glucose level of pomegranate polyphenols administration group has significantly decreased.compared with control group,the Flns and IRI level of model group has significantly increased.compared with model group,the Flns and IRI level of pomegranate polyphenols administration group has significantly has significantly decreased.compared with control group,the level of TC and TG of model group has significantly increased,compared with model group,the level of TC and TG of pomegranate polyphenols administration group has significantly decreased,compared with control group,the level of HDLC of model group has decreased,compared with model group,at some degree,the level HDLC of pomegranate polyphenols administration group has increased,but have no statistical meaning,compared with control group,the level of LDLC of model group has significantly increased,compared with model group,at some degree,the level of LDLC of pomegranate polyphenols administration group has decreased,but have no statistical meaning,compared with control group,the level of ALT、AST of model group has significantly increased,compared with model group,the level of ALT、AST of pomegranate polyphenols administration group has significantly decreased,compared with control group the active of SOD and GSH has significantly decreased,the level of MDA has increased,compared with model group the active of SOD and GSH has significantly increased,the level of MDA has decreased,as well as the fat droplets in the lever cell has significantly decreased compared with model group.Pomegranate polyphenols total polyphenol content is 40.13%,gallic acid 0.005-0.12mg/ml(r=0.9999)in the concentration range of a good linear relationship between peak area,the average recovery 100.25%,RSD=1.64%;Ellagic acid Within 0.01-0.08mg/ml(r=0.9999)concentration range with good linear relationship between peak area,the average recovery 100.59%,RSD=0.87%.Conclusion:The pomegranate polyphenols obviously reduced blood fat levels,reduced aminotransferase and insulin resistance,can protect liver from liver injury.Chromatographic conditions and the method is simple,accurate,which can be used for evaluation of gallic acid and ellagic acid contents in Pomegranate.
