Gastric cancer is a kind of malignant tumors which happens on the gastric epithelial mucosal.Due to the incidence invasion,metastasis characteristics,clinical symptoms,and its low cure rate of gastric cancer,it is one of the major diseases we need to face.The main reason causing high mortality of gastric cancer is the early symptoms of lack of specificity.So it is very difficult for its detection and prevention.In recent years,although the constant cancer gene associated with gastric cancer,tumor suppressor genes and tumor related signaling pathways were found and confirmed,its pathogenesis is not completely clear.Therefore,looking for biological targets for new cancer surveillance and intervention of gastric cancer gastric cancer prevention has become an urgent problem to be solved.Circular RNA?circular RNA,circRNA?is a kind of extensive and diverse endogenous RNA molecules in mammalian cells with gene expression and regulation effect.CircRNA use its microRNA?miRNA?response elements combination of miRNA,in order to block the inhibitory effect of miRNA on the expression of its target,to regulate the expression of other RNA level.CircRNA plays an important role in the disease process of atherosclerosis,nervous system disorders occur,diabetes and cancer,in-depth study of the expression characteristics and function of circRNA could help us better understanding of the pathogenesis of the disease,improve the level of prevention and diagnosis of related diseases.The discovery of circRNA provides a new research field for the study of early diagnosis of cancer.Purpose:Screening of gastric cancer tissue and adjacent tissue expression differences of mRNA and circRNA expression,and analysis of the difference of the expression of circRNA and mRNA,to investigate the expression of circRNA in gastric carcinoma and its mechanism.Method:Selection of 8 cases of surgical removal of the fresh frozen storage of gastric cancer tissue and its corresponding normal tissue adjacent to cancer,total RNA extraction method of Trizol,using NanoDrop ND-1000 to detect whether RNA degradation and the concentration of RNA.Using Agilent microarray technology,analysis of circRNA and mRNA differential expression profile in gastric cancer tissues and adjacent tissues.Using RT-PCR to verify the differential expression of circRNA and mRNA in gastric cancer tissues and adjacent tissues,according to the interaction between circRNA and miRNA,as well as the possible existence of miRNA and mRNA,Analysis of the possible association between circRNA and mRNA in gastric cancer tissues and adjacent normal control tissues.Results:?1?CircRNA expression profile microarray analysis revealed that there are 1285 significant differences in the expression of circRNA,594 circRNA expression was down regulated,and 691 circRNA expression was up-regulated,through the interaction with miRNA,analysis of differentially expressed circRNA,these suggest that circRNA may be closely related to the occurrence of gastric cancer.RT-PCR validation experiments show that the expression of hsacircRN A400071,hsacircRNA000543 and hsacircRNA001959 was consistent with the results of microarray analysis.?2?Using mRNA expression microarray,29112 genes are found in the gastric cancer tissues and adjacent tissues,differentially expressed genes were detected in 5460.Among them,2390 differentially expressed genes were up-regulated and 3070 genes were down regulated.Gene Ontology of differentially expressed genes,There are 1638 genes involved in cellular compoment classification,205 genes involved in molecular function classification,147 genes involved in molecular function classification.Pathway analysis of differentially expressed genes,83 genes were found to be significantly enriched,including 28 up-regulated genes and 55 down regulated genes.RT-PCR experiments show that,the expression of differentially expressed genes MYH9,CD44,ARVCF and so on were in agreement with the results of the microarray analysis.?3?Through the analysis of the correlation between circRNA and mRNA expression profile,69 differentially expressed circRNA were found,may be adsorbed by specific miRNA,regulation of the expression of its target gene mRNA.Conclusion:?1?Differentially expressed circRNA had its corresponding miRNA binding site,through the interaction with miRNA,regulation of the expression of target genes,these circRNA may become a new molecular biomarker for gastric cancer in the future.?2?differentially expressed genes may be involved in the occurrence of gastric cancer in a variety of ways.?3?CD44,CXXC5,MYH9,MALAT1 and other genes through interaction between circRNA and miRNA,miRNA and mRNA,in different ways regulation,interaction,mutual influence,it is possible to have an important influence on the occurrence and development of gastric cancer.
