Objective The aim of the present study was to investigate the relationship between TLR4 expression of COPD rat pulmonary artery smooth muscle cells and its synthesis and secretion function.Methods Animal experiment,chronic obstructive pulmonary disease(COPD)rats were established,HE dyed decision model is established in this paper,Immuno histochemical(IHC)demonstrated that TLR4 expression of pulmonary tissue in the control group and model group.Cellular level test,the PASMCs were isolated from the rat pulmonary artery,TAK-242 the specificity inhibitors of TLR4 block TLR4,groups as follows: the control group,LPS stimulation group,TAK-242 group,LPS+TAK-242 group,the expression of TLR4 in PASMCs was determined with Immunohistochemical(IHC)staining and western blot analysis.Enzyme-linked immuno sorbent assay(ELISA)analysis were used to detect the secretion of inflammatory factors interferon-?(IFN-?)? Interleukin-6(IL-6)? monocyte chemoattractant protein 1(MCP-1)?Transforming growth factor ?1(TGF-?1)?platelet derived growth factor(PDGF)of rat distal PASMCs in each group,followed by a correlation analysis.Results Normal rats lung tissue express TLR4,however TLR4 expression level in model group of COPD rats lung tissue obviously higher than normal group,compared with the control group TLR4 expression of LPS group in pulmonary artery smooth muscle cells increased significantly,LPS induced high TLR4 expression increase the level of PASMCs synthesis and secretion of inflammatory factors IFN-??IL-6?MCP-1?TGF-?1?PDGF evidently,TAK-242 the specificity inhibitors of TLR4 block TLR4 cut the level of PASMCs synthesis and secretion of inflammatory factors IFN-??IL-6?MCP-1?TGF-?1?PDGF evidently,the expression of TLR4 was positively correlated with the synthesis and secretioon of INF-?,IL-6,MCP-1,TGF-?1,PDGF in the PASMCs of COPD rats.The conclusion One,LPS induced can increase the expression of TLR4 in PASMCs;two,TLR4 may participate in regulating the synthesis and secretion of PASMCs.
