With "the Chinese herbal pieces aiding in medicines,raw and processed treating different,accessories processing" characteristics of traditional Chinese medicines processing theory is an important aspect of clinical medicine of trad-itional Chinese medicine,among which processing with salt has a long history and wide application.Chinese medicine processing with salt contributes to brin-ging effective constituents in the kidney,and enhances the effect of nourishing yin to lessen fire.Rhizoma anemarrhenae is the representative of traditional Chinese medicine for "promoting salt into the kidney",Raw Rhizoma Anemarr-henae is good at heat-clearing,fire-purging,increasing the fluid and moistening dryness.Rhizoma anemarrhenae processed can be downward after salt system,with the effects of nourishing yin to lessen fire enhanced.Current researches on processing of Rhizoma Anemarrhenae mainly concentrate on the processing technology and the variation of chemicals.However,how ingredients of Rhizo-ma Anemarrhenae change in vivo before and after processing has few reported.This paper focus on Raw Rhizoma Anemarrhenae and processed one with salt,and take ingredients like the typical flavonoids and saponins as testing indexe s,it carried out content determination,the pharmacokinetics and tissue distribu-tion study.The main research content is as follows:1.Through reading ancient and modern papers,in order to make the theo-retical basis perfect,the historical evolution,the body process,modern processing technology and mechanism of Rhizoma Anemarrhenae was systematically orga-nized.This paper put forward a feasible plan and decided the examining index by means of reviewing the detection principle and application of liquid chro-matography-mass spectrometry technology,which laid the foundation for resear ch.2.By LC-MS,cetonitrile-0.1%formic acid water as mobile phase,content variations of Neo-mangiferin,Mangiferin,Timosaponin BII,Timosaponin B?,Timosaponin A?,Anemarrhenasaponin AII,Timosaponin El,Anemarrhenasaponin I in raw and processed one derived from different regions were simultaneously determined.Results showed that the standard curves were good with the scope,and precision,stability and accuracy were up to the standard.There exist certain differences among different slices of origin content.After processed,content of Neo-mangiferin,Mangiferin,Timosaponin B? and Timosaponin El were reduced,Timosaponin B? rised,meanwhile,Timosaponin AIII,Anemarrhenasaponin AI I had no obvious changes.3.Protein deposition method is used to deal with the rat plasma samples,ginsenosides Rgl and glycyrrhizin choosed as internal standard,using LC-MS/MS method for determination of blood concentration(including Neo-mangiferin,Mangiferin,Timosaponin BII,Timosaponin B? and Timosaponin AIII)in rats which intragastric administrated with extracting solution of medicinal slices of Rhizoma Anemarrhenae and processed.Using DAS 2.0 pharmacokinetic analysis software processing data,discussed the five kinds of compounds in rats in vivo pharmacokinetic characteristics.It turned out that the standard curves were good within the scope,and precision,accuracy and recovery conformed to the relevant requirements.4.Established LC-MS/MS method to test main effective compounds(Neo-mangiferin,Mangiferin,Timosaponin BII,Timosaponin B? and Timosaponin AIII)of rhizoma anemarrhenae in tissue samples of SD rats.After dosing,plasma of SD rats were collected at the appropriate time according to the plasma phar-macokinetic experiment.Using the method to detect absorption phase,distribution phase,eliminate the phase major effect ingredient content of Rhizoma Anema-rrhenae changes in tissue samples,and analysis the distribution of each tissue.The results showed that the content of five compounds in tissues of the same point all exist difference certainly.In summary,this topic has been the object to raw Rhizoma Anemarrhenae and salt-processed Rhizoma Anemarrhenae in our research.Under the guidance of Chiniese medicine theories,we combined multiple modern methods in our research,such as LC-MS/MS Both the research ideas and technologies were innovative.The results could further explain the mechanism of kidney channel tropism of salt-processing Rhizoma Anemarrhenae.It also provides new ideas for the modernization research of TCM.
