QingReXiaoJi Formula's Influence On NF-?B And HIF-1 Alpha Proteins Of Mice With Lewis Lung Transplanted Carcinoma

Objective To detect NF-?B,HIF-1 alpha protein expression of mice with Lewis lung subcutaneous tumor by Immunohistochemical and Western blotting methods.QingReXiaoJi Formula's influence on NF-?B and HIF-1 alpha proteins of Mice with Lewis lung transplanted Carcinoma was explored.Methods 60 C57BL/6J inbred mice with Lewis lung carcinoma xenograft model,the next day.using a random number table method will be divided into five groups,which Qingre Xiaoji square divided into high,medium and low dose groupanother blank control group(saline group,NS group)and the positive control group(cyclophosphamide group,CTX group).The positive control group were treated with intraperitoneal injection of CTX(twice a week),I tumor-bearing mice using gavage,gavage or injection volume were 0.4 ml,1 times a day,for 14 days.Withdrawal the next day,the mice were sacrificed,peel the subcutaneous tumor into the part of the tumor at-80?,ready to Western blotting using;part of the tumor in formalin-fixed-rinse-dehydration transparent-dipDecember-embedded,made of paraffin-embedded tumor tissue to prepare for immunohistochemical method;using immunohistochemical method and Western blot detection of tumor-bearing mice subcutaneous tumor tissue NF-?B,HIF-1 alpha protein expression and so on.Results NF-?B and HIF-1 alpha proteins were significantly decreased in the treatment groups as compared with placebo group(p<0.05).Conclusion QingReXiaoJi formula's efficacy on lowering the expressions of NF-?B and HIF-1 alpha proteins within the tumor tissues may be one of the mechanisms of anti tumor angiogenesis.
