| 1.The purpose of the study:Atrial Fibrillation(AF)is the most common sustained cardiac arrhythmias.The incidence of af with age increasing,over 75 people can reach 10%.The growth of the prevalence of Atrial Fibrillation with diseases such as coronary heart disease,hypertension and heart failure is closely related to the growth of the next 50 years AF will become one of the most popular of cardiovascular disease.This research mainly want to through the animal experiment,the paroxysmal Atrial Fibrillation rats,observe the effectiveness of acupuncture and moxibustion in close hole pretreatment,important index of main monitoring electrocardiogram(ecg)of rats,calculate the duration of Atrial Fibrillation in rats,P wave dispersion degree,experiment in seven days out,take in the rat heart muscle tissue,taking group under the electron microscope observation as well as the similarities and differences of atrial muscle ultrastructure of rats.Through the above indicators to detect acupuncture in paroxysmal Atrial Fibrillation heart rate changes of rat hole pretreatment adjustment is effective.2.Research methods:(1).The experimental equipment,experimental subjects,experimental drug preparation;(2)Groups:the experimental healthy male rats were randomly divided into four groups,after weighing is the blank group,model group,acupuncture group,medication group,10 rats in each group.(3)The establishment of animal model:experimental rats after anesthesia using calcium chloride daily-acetylcholine mixture(Ach 66μg,Cacl2 10mg,on the day of the experimental configuration),through the way of tail intravenous dosing,paroxysmal atrial fibrillation model in rats after the treatment;(4).Experimental methods:blank group rats daily anesthesia yixiang 1 ml saline solution,given intravenously,model group rats injected fresh after anesthesia configuration of calcium chloride solution-acetylcholine,acupuncture group of rats after anesthesia needle closed cavity in retaining needle parallel needle for 10 minutes,30 minutes,30 minutes after injection mold drugs,drug group daily to rats by gastric amiodarone solution(4.5 mg/kg,d),the bid,yixiang intravenous anesthesia model drug.So the experiment for a week,on the eighth day drawing comparison groups of indicators;(5).Be familiar with and master the rat heart muscle based methods;(6).Record and statistics of each group rats af duration,calculating the P wave dispersion of each rat,using the statistical method to calculate each group differences and compare,it is concluded that the experimental results;(7).Accurate records in the process of the experimental rats mortality,and timely analysis of the cause of death in rats.3.The research results:(1)the experimental group after injection of acetylcholine-calcium chloride mixture can cause of paroxysmal atrial fibrillation model of success,success in caudal vein injection building drugs after 1-2 s can appear P wave disappear,small f wave of atrial fibrillation waveform,af duration of 7 s and 20 s.(2)of atrial fibrillation model group rats injected with drug building after 1 day,3 day,5 days,the seventh day of the electrocardiogram(ecg)P wave width and P wave dispersion ratio value after rats compared with normal group,the results are very significant difference(P<0.01).(3)the experiment through 7 days later,the pretreatment of the acupuncture treatment group and drug treatment group rats appear paroxysmal af duration is shorter,the results compared with model group rats have significant difference(P<0.01).(4)groups of rats were injected with building drugs after 7 days,pretreatment of acupuncture group and the use of dragon pharmaceutical group rats paroxysmal af ecg P wave dispersion degree of the concrete numerical value and P wave width of concrete are shorter,compared with atrial fibrillation model group rats after had obvious difference(P<0.01).(5)between groups in the rat heart muscle group under the electron microscope visible atrial fibrillation model group rats myocardial fiber order disorder,muscle wire the length of sarcomere,muscle wire structure is very vague;Obvious swelling of mitochondria,accompanied by crest fusion;Muscle cell nuclear membrane serious shrinkage and nuclear shape is irregular,sarcomere very excited and contraction of muscle fiber and with focal fusion;Visible intercalated disc rupture or intercalated disc fusion.However,pretreatment of acupuncture group and can reach the dragon drug treatment group rats myocardial cells visible only slight interstitial edema,occasionally visible myocardial fibers have owe row order rules,the pathology for the pretreatment of the acupuncture group and can reach the drug therapy group was obviously better than that of AF model group.4,the research conclusion:(1)acupuncture group and can reach the dragon drugs for paroxysmal atrial fibrillation in the rat P wave dispersion ratio value function,shows that acupuncture preconditioning in closed cavity treatment of paroxysmal atrial fibrillation in rats.(2)electron microscope atrial muscle ultrastructure showed that acupuncture group and drug group were superior to atrial fibrillation model group,acupuncture in closing hole to improve the structure of the rat af refactoring is effective. |
