To achieve temperature sensitive release properties, the membrane of thaditional thermo-sensitive liposomes(TTSL)is mainly composed of DPPC,adding 10%lysophospholipids(MSPC),but MSPC has side effects of hemolytic, and single phospholipid is susceptible to drugs,so it is difficult to control the temperature-sensitive release performance in vivo.To overcome the above adverse situation, the technology of composite phospholipids was applied, successfully prepared the NTSL containing total alkaloids of Strychnos L,optimized its preparation, and evaluated the pharmaceutical properties including particle size, potential, transmission electron micrographs, and the drug release properties in PBS with or without rat plasma. In addition, the acute toxicity, pharmacokinetics and tumor targeting properties of TTSL and NTSL were also compared. The results show that, using the composite phospholipids technology, the dosage of MSPC can be reduced by 60 percents, resulting to the improvement of medication security.Meanwhile, compared to TTSL,NTSL can achieve a better thermo-sensitive release performance. And more, the technology of composite phospholipids can achieve the flexibly adjustment of liposomes phase transition temperature, which is on doubt with great significance to the thermo-sensitive liposomes.
