Objective To investigate the diagnostic implications of anti-hTERT autoantibodies in the serum of cancer patients.Methods 1.Induced recombinant bacteria pMAL-c5X-hTERT-n-TB1 to express the fusion protein of hTERT-n.2.A large scale culture of recombinant TB1 for purification of fution proteins hTERT-n by amylose affinity chromatography was performed.The purified protein was then identified by 12%SDS-PAGE and Western blot and quantitated by Coomassie brilliant blue G-250.3.Indirect ELISA was used to detect anti-hTERT-n autoantibodies in the sera of 667 pateints with cancers,120 normal controls and 123 patients with benign diseases.Results The fusion protein hTERT-n was purified with high purity and a concentration of 1.0 mg/ml respectively.The levels of anti-hTERT-n autoantibodies(OD)in serum of patients with cancers are significantly higher than those of normal controls and benigh diseases(P<O.OS).The levels of anti-hTERT-n autoantibodies(OD)in serum of patients with benigh diseases are significantly higher than those of nomal controls(P<O.05).The levels of anti-hTERT-n autoantibodies in serum ot patients with lung cancer,colorectal cancer,breast cancer and ovarian cancer were significantly higher than those of benign diseases(pneumonia).These levels were also higher in pationts with medium or terminal cancer than those in early phase,but there's no different at different ages.Conclusions The levels of anti-hTERT-n autoantibodies in serum of patients with cancers are significantly higher than those of normal controls and benign diseases(pneumonia).The results revealed that anti-hTERT autoantibodies may be a novel biomarker for cancer diagnosis.
