| Antrodia camphorata(M.Zang&C.H.Su)Sheng H.Wu,Leif Ryvarden&T.T.Chang,also named Niuchangchih or "Niu-chang-ku",is a fungus that only grows on the brown heartwood of Cinamomum kanehirae Hayata(Lauraceae)in Taiwan.Antrodia camphorata,known as the "forest ruby",was historically only used in Taiwan by the aborigines as a traditional prescription for the discomforts caused by alcohol drinking or exhaustion.The triterpenoids,polysaccharides,nucleosides and lipids are the main active ingredients.Due to its rarity in the nature and difficulty in culture,currently,Antrodia camphorata is commercially available in the main form of solid-state fermentation in powdered,which has the similar efficacy with wild Antrodia camphorata.In this paper,effective components of Antrodia camphorata were detected in order to evaluate it’s exploitation value.The fingerprinting of Antrodia amphorata based on activity constituent were established,and the pharmacognosy research of fruiting body had been studied.The research mainly includes the following aspects:1.Literature study:Literatures related to Antrodia camphorata up to now are studied thoroughly.2.The pharmacognosy research of the fruiting body:Identified using traditional methods and cryo-sectioning technique,from the medieinal properties,microseopic characteristics,thin-layer chromatography,set up the fruiting body identifieation methods.The localization of polysaccharides and triterpenoids had been done by simple histochemical.Result showed that polysaccharides distributed throughout the fruiting,while the triterpenoids only distributed in the extremity of bacteria tube.3.Eeffective component analysis of Antrodia camphorata powder:The chemical compositions of Antrodia camphorata powder in different batches,different fermentation technology and different manufacturers were analyzed,as well as the capsule and the fruiting body.Additionally,all the components of Ganoderma lucidum had been detected and compared.The nucleosides and nucleobases had been simultaneous analyzed by UPLC-Q-TOF-MS and LC-QTRAP-MS.The volatile components were separated and identified by HSGC-MS.The content of 22 inorganic elements had been detected by ICP-MS.Besides,the free amino acids and combination amino acids were analyzed by pre-column derivatization HPLC.The results showed that all analyzed components had a small difference.These investigated compounds in Antrodia camphorata powder with higher triterpenoid content and Antrodia camphorata capsule represented little distinction comparing with A.camphorata powder.However,remarkable differences were found among the content of each component in A.camphorata sporophore,the A.camphorata powder collected from Guangdong and the Lingzhi samples.4.To establish the fingerprint chromatogram of Antrodia camphorata powder:Simple and reliable UPLC-Q-TOF-MS and HSGC-MS methods has been developed and validated for the fingerprintings of Antrodia camphorata powder.Its common chromatographic peaks of the UPLC-MS and HSGC-MS fingerprints were identified in this experiment,the data were analyzed by Fuzzy Cluster and degree of different fingerprint patterns was calculated and analyzed.It can conduct an overall description and evaluation of the quality of the commercially available products of Antrodia camphorata.For the Antrodia camphorata powder of different batches,the fingerprints of the samples showed closely similar fingerprints.The established fingerprints can quickly distinguish different Antrodia camphorata commodities and Lingzhi samples.5.Study on quality standards of Antrodia camphorata powder:The moisture,ash and water-soluble extract were detected according to the standards stated in Chinese Pharmacopoeia.UV-VIS was used to analyze total polysaccharide and triterpenoid.The content of adenosine was determined by HPLC.Limitation of moisture content,ash,extract,polysaccharide,triterpenoid and adenosine were defined,which could be a quality control standard for commercial Antrodia camphorata powder.Innovations and Features:1.It was the first time to systematically analyze the pharmacognosy research of Antrodia camphorata sporophore,and illustrate its organizational structural characteristics by cryo-sectioning technique.Meanwhile,the distributions of polysaccharide and triterpenoid in Antrodia camphorata were analyzed.2.It was the first time to establish an UPLC-Q-TOF-MS and LC-QTRAP-MS method for simultaneous determination of nucleosides and nucleobases in Antrodia camphorata powder,which can be used as a reliable detection method for the full control of the intrinsic quality of Antrodia camphorata powder.3.To establish the UPLC-Q-TOF-MS and HSGC-MS fingerprint analytical method of Antrodia camphorata powder.A scientific evaluation method was established to find the stability and consistency between different batches.4.To establish the quality of Antrodia camphorata powder scientific evaluation system by quantitative analysis of the various ingredients in combination fingerprint pattern of the overall evaluation.A combination of both can be used for a comprehensive evaluation and full control of the intrinsic quality of Antrodia camphorata powder. |
