Objective To investigate the effect of indometacin on apoptosis of Hela and the suppressive effect of indometacin and cisplatinum on Hela,explore the mechanism of inducing cell apoptosis,and provide theoretical basis for the clinical treatment of indomethacin application to cervical cancer.Methods This study was carried out on culture of human cervical cancer cell line Hela in vitro.The inhibitory rate of indometacin,cisplatin and indometacin combined with cisplatin on Hela was tested by MTT assay.The change of cellular morphology was observed by invert microscope.The nuclear change was observed by fluorescence microscope.The cell cycle phase and apoptosis were analyzed by flow cytometry.The protein expression on apoptosis was detected by Western Blot.Results Indometacin can inhibit the growth of Hela.The inhibitory rate increased accordingly with the increase of drug concentration and action time.Indometacin combined with cisplatin have a stronger effect than monotherapy.The number of cells decreased,cell showed different extent phenomenon of detachment and cell morphology changed significantly from polygon shape to round shape under an optical microscope.Nuclear condensing,fragmenting and condensed staining were observed through fluorescent microscope by Hoechst 33258 dye.Flow cytometry analysis showed that the proportion of cells at G0/G1 phase increased,the proportion of cells at S phase reduced and the proportion of early apoptotic cells raised in a dose-dependent manner.The expression of Bcl-2,Procaspase-9 and Procaspase-3 decreased with the increase of drug concentration while there was no change in the expression of COX-2 and BaxConclusion Indometacin can inhibit the proliferation of Hela and has synergistic effect with cisplatin on the suppressive effect of Hela.Indometacin can arrest the cell cycle in G0/G1 phase and induct apoptosis.Its meachanism is probably involved in the activation of mitochondrial apoptotic pathways.
