Long non-coding RNAs(lncRNAs)are a novel class of mRNA-like transcripts with no protein coding capacity,but with a variety of functions including roles in epigenetics and gene regulation.In recent reports,the aberrant expression of lncRNAs has been associated with human cancers,suggesting a critical role in tumorigenesis.For purposes of investigating the expression profile of lncRNAs involving pancreatic ductal adenocarcinoma(PDAC),as well as the complicated network among lncRNA,microRNA and mRNA,we detected these RNA expression profile in PDAC tissue and paired normal pancreatic tissue by microarray and selected potential competing endogenous RNAs(ceRNAs).Next,Real-time Quantitative PCR(qRT-PCR)was performed to analyze lncRNA expression from 27 patients who underwent pancreatectomy for pancreatic ductal adenocarcinoma.We also investigated the relationship between the expression of n407201 and clinicopathologic characteristics,as well as the prognosis of PDAC patients by statistical methods.Compared with the normal pancreatic tissue,microarry results revealed that there were 570 lncRNAs,47 microRNA and 1409 mRNA aberrant expressed in PDAC tissues(fold change?2,P<0.05,FDR<0.05).Bioinformatics analysis show that there were plentiful lncRNA-microRNA-mRNA network in the PDAC.Furthermore,LncRNA n407201 was found up-regulated in 17/27 patients by qRT-PCR,andsignificant correlated with tumor size,CA199 level and histological differentiation level(P < 0.05).Meanwhile,we confirmed up-regulated LncRNA n407201 may be a novel biomarker for the prognosis of pancreatic cancer.
