On the study of Chinese medicine has a long cultural history,more and more people attention to its treatment of various diseases,but because medicine itself has a multi-component,multi-target,multi-channel work jointly characteristics,and its material base effect is not clear,hindering the modemization,international development of Chinese medicine.Thus,fingerprint technology is widely used in traditional Chinese medicine efficacy of the material basis of the study,and through its inherent pharmacological activity correlations established"profile-effect of Chinese medicine",in order to further reveal the material basis for the efficacy of traditional Chinese medicine to provide a basis.In this study,through the establishment of Cistanche herbs fingerprints and pharmacological activity(mouse uterine weight gain experiment,MTT cell proliferation assay)correlations,and through their analysis reveals Cistanche herbs estrogenic activity may be effective substances foundation.The main research contents and results are as follows:(1)Via Mice Uterotrophic Law and MTT cell proliferation assay on screening different extraction sites and different doses of the Cistanche,the result showed that 95%ethanol extracts of 30 g·kg-1·d-1(crude drug count)as Cistanche estrogen-like effects of the best active site,to lay the foundation for further study of Cistanche herbs estrogenic material basis of the efficacy.(2)Application of HPLC-DAD,fingerprint of 10 origin Cistanche are established,the similarity between the different Cistanche sanples is using by the chromatographic of similarity evaluation(2004),through the study of Cistanche fingerprints show that the best detection wavelength of 254nm,the best elution system of acetonitrile-0.2%formic acid.Provide a basis for the subsequent chemical analysis conducted of the Cistanche herbs and the identification of effective substance authenticity Cistanche herbs.Provide a basis for further deepen the quality control of medicines in cistanche research.(3)Through its HPLC/Q-TOF-MS analysis showed that the mass spectrometry-total ion current chromatogrph are obtained 26 chemical components,total inferred 24 compoundsl,2,4,5,6,8,9,10,11,12,13,14,15,16,17,18,19,20,21,22,23,24,25,26,namely:Genipos idic acid,Decaffeoylacteoside,8-Epiloganic acid,Salidroside,SyringalideA3'-?-L-rhamnopyranoside,Cistanoside A,Kankanoside H1,Echinacoside,Cistanoside F,Cistanoside B,Tubuloside A,Cistanoside C,Osmanthuside B,Acteoside,Cis-acteoside,Isoacteoside,Cistantubuloside Bl,Cistanoside D,Campneoside I,Tubuloside B,Campneoside ?,2'-acetylacteoside,Cistanoside G,Kankanoside E.This conclusion is provide a basis for the further in-depth follow-up study of the efficacy substances of Cistanche herbs.(4)By analysis the correlation of Cistanche herbs spectrum activity relationship,provides a reasonable basis on the efficacy material basis for the Cistanche herbs.By studying on the correlation of 10 fingerprints of origin Cistanche estrogenic activity,draw the following conclusions:There are nine chenical composition were significantly related to the Mice Uterotrophic Law and the corresponding peaks are No.4,5,6,7,8,14,15,16,19;total of ten chemical components associated with the cell proliferation,in which seven components significantly correlated with the cell proliferation and the corresponding peaks are No.3,4,8,12,14,15,19;3 was significantly associated with their constituents.And corresponding Peaks were number of 5,13,18;There are six components were significantly related to the Mice Uterotrophic Law and cell proliferation and the corresponding peak number of 4,5,8,14,15,19.(5)Using D-101 macroporous resin to isolate and purifie the Cistanche herbs,by comparing the fingerprints of different parts of the elution and its corresponding efficacy indicators(uterine factor,MCF-7 cell proliferation rate),indicating that 80%ethanol site for the optimum elution volume fraction of the eluent.Through its analysis of the data showed that isolated and purified Cistanche herbs 17 chemical composition,corresponding peaks are No.3,4,5,8,9,10,13,14,16,17,18,20,21,22,23,25,26,namely:8-Epiloganic acid,Salidroside,Cistanoside A,Kankanoside H1,Echinacoside,Tubuloside A,Cistanoside C,Acteoside,Cis-acteoside,Isoacteoside,Cistanoside D,Campneoside ?,Tubuloside B,Campneoside ?,Cistanoside G,Kankanoside E.The above study suggests that the Cistanche estrogenic activity site,and use HPLC?ESI/MS hyphenated techniques analyze ad identified the Cistanche herbs,have important value for revealing possible estrogen-like effects material basis of the Cistanche,also has some significance for the clinical use of Cistanche.
