Investigation On Adenovirus Infection In Shandong Province

Objective: To investigate the infection status of different adenovirus serotypes in Shandong province in order to find out lower infection serotypes for further adenoviral vector construction in Shandong province.Methods: Five hundreds healthy people aged 18~70 years old from five different cities of Shandong province(Jinan,Qingdao,Weihai,Linyi and Heze)were randomly recruited in the study.Each was taken 5ml of venous blood.The total anti-adenovirus antibody was determined using ELISA.Those with positive results were further tested for individual adenovirus neutralizing antibody.Standard adenovirus strains(serotype 3,5,7,11,26,35,48 and 49)were multiplied in A549 cells and purified with CsCL gradient centrifugation.96-well plates were coated with the adenoviruses.Adenovirus neutral antibody was determined with ELISA using adenovirus coated 96-well plate.Results: The total adenovirus antibody positive rate of the 500 healthy people from five different cities of Shandong Province was 95.2%.There was no significant difference in the total anti-adenovirus antibody between different cities,different age groups and different genders(P>0.05).The positive rate of neutralizing antibodes against adenovirus serotype 3,5,7,11,26,35,48 and 49 were 69.8%,71%,61%,9.4%,2.2%,5.8%,0.8% and 1.6%,respectively.The positive rates of neutralizing antibodies against serotype 11,26,35,48 were significantly lower than those of serotype 3,5 and 7(P<0.05).The positive rates of neutralizing antibody between different cities showed no significant difference(P>0.05);The positive rates of neutralizing antibody was increased along with the age,though the increase was not statistically significant(P>0.05).The median OD values of neutralizing antibodies in 51-70 year old group were significantly lower than the other two groups(P<0.05).There was no significant difference in neutralizing antibodies between different genders(P>0.05).Conclusion: The adenovirus infection rate was high in Shandong province.The infection rates of 26,48 and 49 adenoviruses showed much lower infection level and can be used for constructing adenovirus vector for gene therapy and vaccination in Shandong province.
