ObjectiveIn order to determine the specific mRNA markers for AIDS with spleen-kidney yang syndrome,and provide objective evidence for TCM syndrome differentiation about AIDS.The potential specific mRNA markers of AIDS with spleen-kidney yang deficiency syndrome were checked clinically.MethodsIn this research,we selected 10 significant difference mRNA genes from gene chip screening result,which was AIDS with spleen-kidney yang deficiency syndrome group compared with the normal control group.They are MAP2K7、IFIT1、MAPK14、NLRP6、DOCK4、ABCC2、CXCL9、TBXA2R、STYK1 and KIR2DL4.The samples included patients with HIV/AIDS of spleen-kidney yang deficiency and healthy volunteers were used quantitative real-time PCR(qRT-PCR)technology to detect the expression of mRNA of above 10 genes,with GAPDH as the reference gene were compared,the relative expression of target gene by 2-ΔCt to indicate the value.Through V20.0 statistical software analyzing,we wanted to known the different expression of mRNA genes between HIV with spleen-kidney yang deficiency syndrome group or AIDS with spleen-kidney yang deficiency syndrome group and normal group,and the different expression between HIV with spleen-kidney yang deficiency syndrome group and AIDS with spleen kidney yang deficiency syndrome group.Results1.Compared AIDS with spleen-kidney yang deficiency syndrome group with the normal group:NLRP6 dowm-regulated in AIDS with spleen-kidney yang deficiency syndrome group,which consistented with the microarray result,and the difference was statistically significant(P<0.05);MAP2K7 and KIR2DL4 up-regulated in AIDS with spleen-kidney yang deficiency syndrome group,which consistented with the microarray result,and the difference was statistically significant(P<0.05);TBXA2R up-regulated in AIDS with spleen-kidney yang deficiency syndrome group,which consistented with the microarray result,but the difference was not statistically significant(P>0.06);CXCL9 up-regulated in AIDS with spleen-kidney yang deficiency syndrome group,which was inconsistent with the microarray result,but the difference was statistically significant(P<0.05);ABCC2 down-regulated in AIDS with spleen-kidney yang deficiency syndrome group,consistent with the microarray result,but the difference was not statistically significant(P>0.05).The expression of IFIT1,DOCK4,STYK1,MAPK14 mRNA in AIDS with spleen-kidney yang deficiency syndrome group was up-regulated,which was inconsistent with the microarray result,and the difference was not statistically significant(P>0.05).2.Compared HIV with spleen-kidney yang deficiency syndrome group and normal group:NLRP6 and ABCC2 down-regulated in HIV withspleen-kidney yang deficiency syndrome group,which consistented with the microarray result,and the difference was statistically significant(P<0.05);MAP2K7 and KIR2DL4 up-regulated in HIV with spleen-kidney yang deficiency syndrome group,which consistented with the microarray result,and the difference was statistically significant(P<0.05);TBXA2R up-regulated in HIV with spleen-kidney yang deficiency syndrome group,which consistented with the microarray result,but the difference was not statistically significant(P>0.05);CXCL9 up-regulated in HIV with spleen-kidney yang deficiency syndrome group,which was inconsistent.with the microarray result,but the difference was statistically significant(P<0.05);DOCK4,MAPK14,STYK1 and IFIT1 mRNA down-regulated in HIV with spleen-kidney yang deficiency syndrome group,which was inconsistent with the microarray result,and the difference was not statistically significant(P>0.05).3.Compared AIDS with spleen-kidney yang deficiency syndrome group with HIV with spleen-kidney yang deficiency syndrome group:the expression of MAP2K7 in the two groups was statistically significant(P<0.05),the remaining nine genes in the two groups were not statistically insignificant(P>0.05).Conclusions1.This research concluded that the specific expression of mRNAs in HIV/AIDS with spleen-kidney yang deficiency syndrome are KIR2DL4,NLRP6and MAP2K7.Those mRNAs may regulate inflammation,immune response,cell apoptosis,etc through the NK cells,the NF-κB signaling pathway and MAPK signal pathways.2.The specific expression of mRNA in HIV with spleen-kidney yang syndrome is ABCC2,ABCC2 may transport the inflammatory signaling molecules such as LTB4,INF-γ and TNF-α to mediate immune reaction.
