Mori cortex comes from the root bark of Morus alba.L,which is a medicinal plant of traditional Chinese medicine call "Sangbaipi".In ancient China,Mori cotex wai used for relieving asthma water swelling and purging lung.There are a lot of active constituent in mori cortex,including flavonoids,alkaloids,stilbenes and polysaccharide compounds.Modern pharmacological studies show that extraction of mori cortex lowering blood pressure,lowering blood sugar level,anti-tumor and treatment of edema and other effects,the most active part of mori cortex is flavonoids extraction.The flavonoids of Morus alba L.Are all connected with the structure of the isopentene group,and the activity of flavonoids is enhanced by the addition of the isopentene group.study of mori cortex show thatflavonoids contains Diels-Alder compounds.structure of such compounds which is chalcone or its derivatives reacting "4+2" cycloaddition reaction.Diels-Alder compounds is connected with isopentene group.Some researchs show that pharmacological effects of Diels-Alder compounds is similar to isopentene flavonoids of Morus alba L.This study is determined to investigate the content of main effective components of total flavonoids in mori cortex and set up a rapid,simple and reliable method for the identification of flavonoids extraction from Morus alba L.Discussing thestability and the pharmacological effect of flavonoids extraction of mori cortex.1.Investigation of the content in lavonoids extraction of mori cortex.Using high performance liquid chromatography,with sanggenon C and D and morusin as the reference.In order to determine the content of sanggenon C and D and cortexmori among total flavonoids extraction.2.Rapid identification of total flavonoids from mori cortex.The extraction was separate and identify in ESI negative mode by ultra performance liquid chromatography coupled with quadrupole tiem-of-flight mass spectrometry(UPLC/Q-TOFMS)technique.According to the experimental result,11 flavonoids,including 7 Diels-Alder and 4 prenylated flavonoids,were separated and identified by UPLC/Q-TOF MS based on precise molecular weight.The loss of H2 and H2O was characterized here for the first time and their fragmentation behavior upon electrospray ionization has been discussed.3.Stability investigation of mori cortexDetecting the changes of mori cortex indifferent temperature,humidity and light,especially the contents of sanggenon C,D and morusin.After accelerated test and long-term stability test,Evaluating the mori cortex’s term of validity.4.Effects of flavonoids from Morus alba L.and rutin on T2DM rats.STZ injection combined high fat feed,in order to create type 2 diabetes mellitus rats.Investigating the pharmacological effect of flavonoids exaction of mori cortex only,rutin only,or the combining flavonoids and rutin.5.Effects of flavonoids from Morus alba L.on T2DM rats.STZ injection combined high fat feed,in order to create type 2 diabetes mellitus rats.Investigating the pharmacological effect of flavonoids exaction of mori cortex between preventing group and treating group.
