Objective: To detect the expression of SIRT3,PI3 K,Beclin1 and LC3-? in hepatocellular carcinoma and adjacent tissues,and to investgate the expression of SIRT3 in HCC through PI3K/Akt/mTOR pathways to the possible role of autophagy in cells.Method:1.28 specimens from patients who were diagnosised with HCC and received operation in Hunan Provincial People's Hospital from October 2015 to October 2016 were put in the study.Immunohistochemistry and Western-blotting were used to detect the expression of SIRT3 in cancer and adjacent tissues,and the correlation was analyzed statistically.2.The expressions of PI3 K,Beclin1 and LC3-? in cancer and adjacent tissues were detected by immunohistochemistry and Western-blotting.The results were analyzed to detect the relationship between the above factors and the possible effect of SIRT3 on autophagy through PI3K/Akt/mTOR pathway.Results:1.The positive expression rate and protein expression of SIRT3 protein in hepatocellular carcinoma were lower than those in adjacent tissues(P <0.05).2.There was no significant difference in the expression level of SIRT3 between hepatocellular carcinoma and the age,sex and tumor size(P> 0.05),but there was significant difference between the degree of tumor differentiation and clinical stage(P<0.05).3.The positive expression rate and protein expression of PI3 K protein in hepatocellular carcinoma were higher than those in adjacent tissues(P <0.05).4.The positive expression rate and protein expression of Beclin1 and LC3-? were lower than those of adjacent tissues(P <0.05).Conclusion:1.The low level of SIRT3 in hepatocellular carcinoma suggested that SIRT3 may affect the level of autophagy in hepatocellular carcinoma,and play a role in the development of hepatocellular carcinoma.2.The low level of SIRT3 may regulate the autophagy in hepatocellular carcinoma through PI3K/Akt/mTOR pathway.
