Objective: Talin-1 is a cytoskeletal protein,which is a key factor among the extracellular matrix-integrin-cytoskeleton proteins,and plays an important role in tumourgenesis,migration and metastasis in several malignant tumors.However,it is not clear whether it is involved in the aggressiveness of breast cancer.The expression status and distribution of Talin-1 protein in 206 breast cancer specimens was analyzed using immunohistochemistry staining.Evaluating the relationship between Talin-1 protein and clinicopathological parameters such as age,tumor stage,lymph metastasis and ER,PR.The aim of this study is to investigate the effect of Talin1 to local invasion and metastasis in breast cancer.It can provide references for early screening,clinical diagnosis,comprehensive treatment and prognosis of breast cancer.Methods: 1.Using immunohistochemistry staining to analyze the expression of Talin-1 protein in 206 breast cancer specimens that was diagnosed by pathology.Collecting the clinical pathological parameters of breast cancer patients,including age,tumor stage and lymph node metastasis to analyze the correlation between the expression level of Talin-1 and the prognosis of these patients.2.The inclusion criteria were as follows:(1)curative operations were performed;(2)resected specimens were pathologically examined;(3)more than ten lymph nodes were pathologically examined after operation;and(4)complete medical records were available.3.In this study,The Chi-square test or Fisher's exact test were used to analyze the relationships between Talin-1 protein expression and clinical characteristics.The Kaplan-Meier method was used to estimate DMFS,and the differences were compared using the log-rank test.All statistical analyses were performed using SPSS 19.0 software(SPSS,Chicago,IL,USA);two-tailed P-values < 0.05 were considered significant.Results: Talin-1 was mainly distributed in the cytoplasm of breast cells,and Talin-1 protein was expressed positively in 42.71% of the 206 cases examined.The expression of Talin-1 protein was related to lymph node metastasis and HER-2(P <0.05).On the other hand,they were not correlated with tumor size,ER,PR,Ki-67 antigen.(P >0.05).Kaplan-Meier survival curves showed that survival rates of patients with positive expression of Talin-1were significantly lower than those with negative expression.Conclusion: 1.Talin-1 expression was significantly associated with lymph node status whlie absence of Her-2 expression.2.Talin-1 expression was associated with a significantly worse overall survival.3.The expression of Talin-1 in breast cancer is an independent negative prognostic factor.
