Objective:To establish a method for hydroxyl radial(·OH)detection in micro-sample and paper test,and the methods have antioxidant properties of antioxidant activity.Methods:⑴Mn2+-H2O2-EBT could be an effective and feasible method to determine the antioxidant activity of a substance.The system was(NaB4O7-NaOH)300μL、EBT120μL、Mn2+300μL、H2O2150μL、add distilled water to 2mL,then detected the absorbance at 560nm.This study was going to test the feasibility of this method.The method of micro-sample aniti hydroxyl radical(·OH)active real-time detection of the feasibility to ascorbic acid for comparison.⑵The principle of Fenton reaction,UV-Vis Spectrophotometric method to optimize the hydroxy naphthol blue(HNB)、calconcarboxylic acid(NANA)、4-(2-pyridylazo)-jane resorcinol(PAR)、alizarin red、dibenzofurans bitter hydrazine radical(DPPH·)、Black T(EBT)、5-Br-PSAA such as pyridine azo compounds.such as wavelength、the largest concentration of chromogenic agent、pH condition such as metal ions and[H2O2],and the determinnation of hydroxyl free radical production.According to the optimized conditions of chromogenic agent on the in vitro experiment to strip,further optimize test reaction conditions,test paper insert after 2min,after natural drying and H2O2,observing the color change of the test.In order to verify the feasibility of this method,vitaminC、ferurine and mannitol were compared.Results:⑴The results show that,The ratio of scavenging on hydroxyl radical(·OH)of those samples bleeding sap of Xinjiang grape tree、Honeysuckle tea、Luobuma tea、Jasmine tea、Bamboo leaf tea has a strong hydroxyl radical(·OH)scavenging,is a good nature resistance to oxidation free radical remover.⑵By ultraviolet-visible spectrometry to examine and optimization of seven kinds of chromogenic reagents,and the optimized conditions of the moving strip,the resulting in vitro reaction and test results are consistent,the test paper method can detect have antioxidant properties of hydroxyl free radical ability.Conclusion:⑴The results show that this method can be used in online detection.And it is able to analyze micro-sample in series.In this test,established detection method of Mn2+-H2O2 system can generate strong hydroxyl free radical(·OH),can be used for antioxidants clearance determination of hydroxyl radical(·OH)activities.⑵The establish test detection method is simple、convenient、fast,can be used for the antioxidants clearance on the activity of hydroxyl radical(·OH)rapid detection method.
