Objective:TO study differential expression of TOPK/PBK response in children with malignant lymphoma and lymph node hyperplasia tissues;To discuss the differential expression of TOPK/PBK in different types of lymphoma,and the relationship between TOPK and clinical pathological parameters was analyzed by pathological and clinical data.Methods:80 specimens of children with malignant lymphoma and 20specimens of reactive hyperplasia of lymph node paraffin were collected from Hunan Provincial People,s Hospital.the expression of TOPK/PBK in all specimens was detected by Immunohistochemistry;the expression of EBER was detected by in situ hybridization in lymphoma specimen;The LMP1 expression of EBER positive lymphoma was detected by immunohistochemical method;The relationship between 80cases of malignant lymphoma in children with TOPK/PBK expression and the pathological types?clinical features and prognosis of EBV infection was analyzed by statistical method.Results:1.80 cases of male to female ratio was 3:1;the age of onset was more than 6 years old;the whole group of lintralymph node onset was51.25%,Out of lymph node onset was 48.75%;the expression of TOPK was not related to age,gender,location of onset;2.The positive rate of TOPK in lymphoma specimens were higher than that of reactive hyperplasia of lymph node specimens,the difference was statistically significan(?2=4.967,P=0.026);The expression of TOPK had no diffrences between HL and NHL.And There was no difference in TOPK expression between the two subtypes of HL;The positive rate of TOPK in lymphoblastic lymphoma was higher than those of the mature B cell lymphoma and T/NK cell lymphoma,the difference was statistically significant(P=0.008 and 0.009,respectively);The expression of TOPK between the mature B cell lymphoma and the mature T/NK cell lymphoma showed no difference;3.The expression of TOPK was positively related to Ki67,and the difference was statistically significant(r=0.295,P=0.013);4.The positive rate of TOPK in LDH increased group was higher than that in the normal group,the difference was statistically significant(?2=6.267,P=0.012);5.The positive rate of TOPK in the stage III+IV group is higher than that of the stage I+II group,the difference was statistically significant(?~2=4.715,P=0.030);6.The positive rate of TOPK in EBER positive lymphoma was higher than that of EBER negative lymphoma,the difference was statistically significant(?~2=4.925,P=0.026);The positive rate of TOPK in EBER positive LMP1 positive lymphoma was higher than that of LMP1negative lymphoma,the difference was statistically significant(?~2=5.712,P=0.017);7.TOPK positive lymphoma patients five-year survival rate is lower than the TOPK negative group,the difference was statistically significant(?~2=7.701,P=0.006).Conclusion:1.The expression of TOPK was up-regulated in children with malignant lymphoma;2.The expression level of TOPK was correlated with the pathological types of malignant lymphoma in children,the more the tumor is malignant,the more it may express;3.The expression of TOPK was positively related to the expression of Ki-67 in malignant lymphoma;4.The expression level of TOPK was higher in LDH rising lymphoma and in advanced malignant lymphoma;5.TOPK is associated with EBV infection in children with malignant lymphoma;6.The expression level of TOPK can be used as an independent prognostic factor in children with malignant lymphoma.
