| Objective:Differential expression and functional characterization of etastasis-related miR-29C in the epithelial ovarian cancerMethods:We diagnosed 120 epithelial ovarian cancer patients from March 2012 to November 2014 in the Department of Gynecological Cancer of Xiangyang Central Hospital.All patients underwent debulking and were diagnosed with EOC based on a histopathological evaluation.We sampled 90 cases of disease ovarian tissue(DC)and 30 cases of normal ovarian tissue(NC)from 120 epithelial ovarian cancer patients,which signed informed consent and contained 30 patients with non-metastasis(NM),30 patients with lymphatic metastasis(LM),30 patients with distant metastasis(DM).Test miR-29c expression in patients with normal ovarian tissues,non-metastasis epithelial ovarian cancer,with lymphatic metastasis of epithelial ovarian cancer and distant metastasis of epithelial ovarian cancer.Focus on the different in the proliferation,apoptosis and PTEN expression of SKOV3 cell line after transfection miR29c-mimics and inhibitor to analyze the effect of miR-29c on the growth and invasion of ovarian cancer.Result:By real time PCR,we confirmed the expression of miR-29c in human epithelial ovarian cancer tissues and cell lines decreased significantly.We also found that miR-29c can cause the inhibition of cell proliferation,migration and invasion ability,and enhance the activity of miR-29c increased the apoptosis rate.In addition,we found that the expression of miR-29c was positively correlated with the expression of tumor suppressor gene PTEN in ovarian cancer.Conclusion:The expression of miR-29c in ovarian cancer tissues was lower than that in normal ovarian tissues.The miR-29c in the occurrence and development of epithelial ovarian cancer may be through the regulation of PTEN gene expression play an inhibitory effect.The results show that the expression of miRNA-29c play an important role in the process of metastasis of epithelial ovarian cancer. |
