Objective:By studying the gene expression of Langchuangding on GRP of CD4+T Cells in SLE under intervention of IL2 and IL4,from the different sensitivity of Systemic Lupus Erythematosus patients,so to explain the Langchuangding takes effect by regulating and comtrol GR?.So it can provide a reliable basis for TCM treatment of SLE.Methods:Screening GC resistant and sensitivity SLE patients who consistent with the selectedstandard to collect the peripheral blood and sort CD4+T cells immediately.CD4+T cells were randomly divided into 4 groups:High dose surum+DXM group,Moderate dose surum+DXM group,Low dose surum+DXM group,Blank control group.The concentration of serum would be 15%,10%,5%respectively.Add the solution of CCK8,after 4h,read the OD values at the 450nm.Than theCD4+T cells were planked and randomly divided into 3 groups:Blank control group.(added blank serum),DXM group:(added blank serum+lOuMol/ml DXM,DXM+surum group:(added serum mixed with Chinese medicine+10uMol/ml DXM.The concentration wasbased on the results of the previous experiment.Add the50U/ml IL2 or 50U/ml IL4.Respectively..The expression of GR? was detected by flow cytometry.Result:The viability of SLE CD4+treated by high,moderate and low dose surum+DXM group was significantly lower than the blanck group(P<0.05).There are the result of cell viabilityspecificly:Low dose surum+DXM group>Moderate dose surum+DXM group>High dose surum+DXM group.After the detection of GC-sensitive SLE CD4+T cellsGRp byflow cytometry,we could find that,the expression of GR?with the DXM+serum is lower than DXM group and CK group(P<0.01).After the detection of GC-resistance SLE CD4+T cells,it showed that he expression of GRP with the DXM+serum is lower than DXM group and CK group(P<0.01).The expression of GR? in DXM group is higher than the CK group stimulated by IL-4(P<0.05).Conclusion:1.Langchaungding can reduce the expression of GR? of GC sensitivity SLE CD4+T cells.2.Langchuangding can reduce the expression of GR? of GC resistance SLE CD4+T cells to increase the GC sensitivity,and help to decrease the dosage of GC.3.Langchaungding can help to ruduce GC dosage by ruduceing the expression of GR?.And it may be an important means to develop the synergy mechanism between Langchaungding and GC.
