Breast cancer is a malignant tumor that occurs in the glandular epithelial tissue.The incidence of breast cancer in women is much higher than that of men.Breast cancer seriously endanger women's physical and mental health,since the 20th century,80 years,breast cancer research has gradually become an international medical research hot spot.Breast cancer is a class of highly heterogeneous malignant tumors,breast cancer is generally divided into luminal type A,luminal B type,HER-2(+)type,basal-like type.According to the expression of estrogen receptor(ER),progesterone receptor(PR)and human epidermal growth factor receptor 2(HER-2)in breast cancer cells and tissues,luminal type A was characterized by ER positive,HER2 negative and/or PR-negative;luminal B is characterized by ER-positive,HER2-positive and/or PR-positive;HER-2(+)is characterized by ER-negative,PR-negative and HER2-positive.In addition,it is generally believed that basal-ER,PR,HER2 were negative,that is,triple negative breast cancer(ER-,PR-,HER2-).Nur77/TR3/NGFI-B/NR4A1 is an important member of the nuclear receptor family.So far,there is no clear endogenous ligand,it is also known as orphan nuclear receptor.Nur77 is abnormal expression in breast tissue and breast cancer cell lines.This study found that knockdown of Nur77 expression in ER-positive MCF-7 cells inhibited the expression of GREB1 and TFF1,which is induced by estrogen,and regulated by ER binding enhancer.In addition,knocking down the expression of Nur77 inhibited estrogen-dependent growth of cell proliferation,and greatly affected estrogen-induced cyclinDl expression and cycle progression.We further found that estrogen(E2)can affect the interaction of ERa with Nur77.In this study,the role of Nur77 in triple-negative breast cancer cells was also studied.Different cytokines induced Nur77 expression and affected the localization of Nur77 cells.In addition,small molecules that targeted Nur77 affect EGF-induced ERK activation.
