| Jujube pigment is a kind of water soluble pigment containing polyphenols and flavonoids.It not only has a good scavenging ability to DPPH radical,hydroxyl radical and superoxide anion radical,but also has a certain inhibitory effect on lipid oxidation and K562 cell,and it can obviously improve the injury of endothelial cell induced by H2O2.In recent years,plant polyphenols become the research focus because of its high antioxidant,anti-radiation,anti-tumor and other biological functions,but the study on polyphenol in jujube pigment has not been reported.Polyphenol in jujube pigment is a main active ingredient in jujube pigment,it studied the protective effects of polyphenol of jujube pigment on oxidative stress in brain,kidney and liver in mice by the models of mice normobaric hypoxia,KBrO3 induced oxidative stress kidney injury in mice and restraint stress induced liver injury animal test in this project.The results were as follows:(1)The effect of polyphenol in jujube pigment on oxygen consumption in 15 minutes were observed in mice suffered from normobaric hypoxia,and the superoxide dismutase(SOD)、malondialdehyde(MDA)in serum,the glutathione peroxidase(GSH-PX)、lactate dehydrogenase(LDH)activities,malondialdehyde(MDA)、NO content and the level of total antioxidant capacity(T-AOC)were determined by ELISA kit.The results showed that:the oxygen consumption was decreased significantly,the SOD in serum、GSH-PX and T-AOC in brain were increased significantly,and the MDA content in serum、the NO and LDH in brain were decreased significantly in mice suffered from normobaric hypoxia by polyphenol in jujube pigment.This result indicated that polyphenol in jujube pigment could improve the effect of oxidative stress on brain injury by improving the oxygen utilization rate of hypoxia mice,enhancing the antioxidant enzyme activity and reducing the generation of peroxide.(2)Building the model of KBrO3 induced oxidative stress kidney injury by 6 h intraperitoneal injection.The protective effects of polyphenol in jujube pigment on oxidative stress induced kidney injury were evaluated by observing the pathological changes under light microscope and determining the content of Urea nitrogen(BUN)and creatinine(Cr)in serum,GSH-PX、Glutathione(GSH)and MDA in kidney.The results showed that:It had a good repair effect on pathological damage.Meanwhile the contents of BUN and Cr in serum and MDA in kidney were decreased significantly,and the activity of GSH-PX in kidney were increased significantly in mice suffered from kidney injury by polyphenol in jujube pigment.This results indicated that polyphenol in jujube pigment could improve KBrO3 induced oxidative stress kidney injury in mice by improving the content of antioxidant substances and oxidation state in body.(3)Building the model of oxidative stress induced liver injury in mice by 18 h restraint load.The effect of pathological morphology of liver tissue was observed by HE staining.The effect of polyphenol in jujube pigment on the peroxidation of mice was evaluated by dete(?)the activities of Glutamic pyruvic transaminase(ALT)、SOD in.serum and liver tissue,and determining the activities of GSH-PX,the contents of MDA and NO.And the effect of mitochondrial function in mice was observed by Gianas green B staining.The results showed that:The pathological damage of liver tissue caused by restraint was improved by polyphenol in jujube pigment,the activities of ALT and the contents of MDA,NO in liver tissue was decreased significantly,and the activities of SOD and GSH-PX in serum and liver tissue were increased significantly.The phenomenon of mitochondrial dysfunction induced by restraint load was significantly reversed.This results indicated that polyphenol in jujube pigment is possible to achieve the protective effect of oxidative stress induced liver injury by enhancing the antioxidant enzyme activities,decreasing the degree of lipid peroxidation and protecting the function of mitochondria. |
