| Objective: To observe the therapeutic effect of Xiaochengqi Decoction on postoperative ileus in rats.The relationship between the mechanism of action and the NF-kappa B / I kappa B signal pathway was preliminarily discussed.To provide a theoretical basis for clinical application of Xiaochengqi Decoction to improve postoperative ileus.Methods: Thirty-two adult male rats were randomly divided into four groups:sham operation group,POI model group,Xiaochengqi Decoction low-dose intervention group,and Xiaochengqi Decoction high-dose intervention group.Rat POI model was constructed by mechanical intestinal control.Small intestine advancing rate and HE staining were to evaluate efficacy of Xiaochengqi Decoction on POI.Detection of TNF-α,IL-6 and IL-1β in rat Serum by ELISA.q PCR and Western blot were used to detect the mRNA and protein expression of P65,IκBα and TNF-α in rat intestine.Results:1.Compared with the POI model group,the Xiaochengqi Decoction high-dose intervention group can significantly increase the rat intestinal propulsion rate(p<0.05);2.Xiaochengqi Decoction intervention group can improve POI-induced pathological injury to varying degrees;3.Xiaochengqi Decoction intervention group can significantly inhibit the release of inflammatory cytokines TNF-α,IL-6 and IL-1β in POI rats(p<0.05);4.Xiaochengqi Decoction low dose intervention group can significantly inhibit the mRNA and protein expression of TNF-α,and can significantly increase the mRNA and protein expression of IκBα;5.Xiaochengqi Decoction high dose intervention group can significantly inhibit the mRNA and protein expression of P65 and TNF-α(p <0.05),and can significantly increase the mRNA and protein expression of IκBα(p <0.05);6.The high-dose group of Xiaochengqi Decoction had a better effect onimproving POI in rats.Conclusion: Xiaochengqi Decoction can effectively improve the POI in rats,and the effect of high-dose intervention is better.The mechanism may be through the regulation of NF-κB/IκB signaling pathway to regulate the release of inflammatory factors such as TNF-α,IL-6 and IL-1β. |
