Objective: In this study,human epidermal growth factor of 180 breast cancer specimens was studied by fluorescence in situ hybridization(FISH)and silver-enhanced in situ hybridization(SISH).The human epidermal growth factor receptor 2(HER2)gene was detected and the gene expression results were analyzed.The coincidence and difference of HER2 gene detection results in two invasive breast cancers were compared to provide the final diagnosis and clinical treatment.Better scientific basis.Methods: From the case information of all breast cancer patients in the electronic archive bank of the Department of Clinical Pathology of the First Affiliated Hospital of Wannan Medical College,the screening time segment was from January 2016 to December 2017.The histological diagnosis was breast A total of 233 cases of infiltrating ductal carcinomas were examined for HER2 protein expression by immunohistochemistry(IHC).The results were negative(0/1+)in 80 cases,indeterminate(2+)in 90 cases,and positive in(3).+)63 cases.Sixty samples were randomly selected from 0/1+,2+,and 3+ cases for FISH and SISH,respectively,according to the 2013 Breast Cancer Association/American College of Pathologists Association(ASCO/CAP)."Guide to cancer HER2 testing" analysis of the results and statistical analysis of the correlation between the two detection methods to analyze the reasons for the differences.In this experiment,the HER2 gene FISH detection kit for breast cancer was purchased from Anping,Guangdong,and the SISH kit was selected from Ventana,a company owned by Roche.The instruments used in the experiment were Bench Mark XT automatic immunohistochemical instrument from Roche Ventana,the BOND-MAX automated immunohistochemical instrument from Leica,Germany,and the Olympus BX51 fluorescence microscope.RESULTS: The results of immunohistochemistry were 0/1+,2+,and 3+ in 60 cases respectively.FISH and SISH were performed at the same time.The results were satisfactory.1.FISH test results: In the 60 cases with HER2 protein negative(0/1+)immunohistochemistry,HER2 gene was not amplified by FISH;the expression of HER2 protein in immunohistochemistry was indeterminate(2+)in 60 cases.,FISH HER2 gene detection results in HER2 gene amplification in 20 cases,HER2 gene not amplified in 38 cases,HER2 gene amplification indefinite in 2 cases;60 cases of immunohistochemistry,HER2 protein expression positive(3+)FISH gene results Both are amplified.2.SISH test results: In immunohistochemical HER2 protein negative(0/1+)60 cases,HER2 gene was not amplified by SISH;immunohistochemical HER2 protein expression was indeterminate(2+)in 60 cases,SISH The results of HER2 gene amplification in 18 cases,40 cases of gene amplification,2 cases of indeterminate gene amplification status;immunohistochemistry results were positive(3+)in 60 cases,59 cases of HER2 gene amplification were detected by SISH.One case of HER2 gene amplification was indeterminate.Compared with the results of FISH and SISH in 180 patients,in the 60 cases with negative immunohistochemical HER2 protein expression(0/1+),no amplification was detected by FISH and SISH.The coincidence rate of the two methods was 100%;immunohistochemistry Indeterminate(2+)HER2 protein expression in 2 out of 60 cases,2 cases were detected without amplification by SISH and gene amplification by FISH.The coincidence rate of the two methods was 96.7%;immunohistochemical HER2 protein expression was positive(3+)Among the 60 cases,1 case of SISH detection gene amplification status was not clear but FISH detection was gene amplification.The coincidence rate of the two methods was 98.3%.After statistical analysis,the results of the two methods are highly consistent.Conclusion: For the two in situ hybridization methods using FISH and SISH,the expression of 60 immunohistochemical HER-2 protein-negative(0/1+)and 60 immunohistochemical HER2 proteins was indeterminate(2+)and 60 respectively.Cases with positive immunohistochemical HER-2 protein expression(3+)were tested and the results analyzed.It was concluded that the coincidence rates of FISH and SISH methods were highly consistent,but there were also certain differences.The immunohistochemical HER2 protein was indeterminate(2+)and positive(3+)when FISH and SISH were performed.
