ObjectiveTo investigate the expression and clinical significance of TM4SF5 in invasive breast cancer tissues,to analyze the relationship between the expression of TM4SF5 and clinicopathological factors in invasive breast cancer,and to elucidate the relationship between TM4SF5 and survival and prognosis of invasive breast cancer.Methods1.The paraffin sections of 180 cases of invasive breast cancer tissues and adjacent normal breast tissues were collected.The expression of TM4SF5 protein was detected by immunohistochemistry in invasive breast cancer tissues and corresponding adjacent normal breast tissues.The expression level of TM4SF5 was analyzed and analyzed.Correlations of clinical pathological features such as age,tumor size,histological grade,lymph node metastasis,TNM stage,ER,PR,HER2,and molecular typing in patients with invasive breast cancer,and follow-up of 180 patients of breast cancer patients,correlation analysis is used to analyze the expression of TM4SF5 in patients with invasive breast cancer prognosis by Kaplan-Meier;2.We collected 60 cases of fresh invasive breast cancer tissue and adjacentnormal tissue samples.The relative expression levels of TM4SF5 protein in invasive breast cancer tissues and adjacent normal tissues were determined by Western blot and the differences were compared.3.We collected 60 cases of fresh invasive breast cancer tissue and adjacent normal tissue samples,and detected the relative expression levels of TM4SF5 mRNA in invasive breast cancer tissues and adjacent normal tissues by real-time quantitative PCR(qRT-PCR)and compared the differences.Results1.The results of immunohistochemistry showed that the expression of TM4SF5 protein is mainly in cell membrane and cytoplasm in invasive breast cancer,and it is expressed in a small amount in nucleus.The positive expression rate of TM4SF5 protein in invasive breast cancer(57.8%)was higher than that in adjacent breast tissues(P < 0.05).there are differences between the two groups;The expression of TM4SF5 protein was not related to age,tumor size and PR status(P > 0.05),and was related to the degree of differentiation,lymph node metastasis,clinical TNM stage,ER status,Her-2 status and molecular typing(P < 0.05).The positive expression rate of TM4SF5 protein in different types of breast cancer was different,the difference was statistically significant(P<0.05).The positive expression rate of TM4SF5 protein in basal-like breast cancer tissues(79.4%)was higher than that of Luminal A(46.8%),Luminal B type(53.7%)and Her-2 over expression type(67.9%);there were significant differences between Luminal A type and basal-like breast cancer tissues(P<0.05)as well as Luminal B type and basal-like(P<0.05),There is no statistical difference between the other two.(P>0.05).The results of Kaplan-Meier single factor survival analysis showed that the 5-year overall survival rate of invasive breast cancer patients with positive TM4SF5 protein was 79.8%,which was significantly lower than the 5-year overall survival rate of invasive breast cancer patients with TM4SF5 protein negative group(90.8%).the difference was statistically significant.(P<0.05);2.Western blot showed that the relative expression level of TM4SF5 protein ininvasive breast cancer tissues(0.607±0.154)was higher than that in adjacent normal tissues(0.105±0.028);the relative expression of TM4SF5 protein in invasive breast cancer tissues and adjacent normal tissues.There was a statistically significant difference in the relative levels of TM4SF5 protein in invasive breast cancer tissues and adjacent normal tissues.(t=5.238,P < 0.05);3.Real-time PCR showed that TM4SF5 mRNA in invasive breast cancer tissues,the relative expression level(5.471±2.084)higher than that in adjacent normal tissues(1.022±0.576);The relative expression levels of TM4SF5 mRNA in invasive breast cancer tissues and adjacent normal tissues were statistically significant different.(t=8.023,P < 0.05)?Conclusions1.TM4SF5 is highly expressed in invasive breast cancer tissues,suggesting that TM4SF1 may play an important role in the development and progression of invasive breast cancer.TM4SF5 may be an important molecular marker of invasive breast cancer and a potential target for invasive breast cancer treatment.2.TM4SF5 may be associated with poor survival prognosis in patients with invasive breast cancer.
