| Objective:1、Identification of bile acids in five animal biles including snake,pig,cattle,sheep and lapin by thin layer chromatography(TLC),polymerase chain reaction(PCR)and LC-MS to solve the problem of adulteration.2、HPLC-CAD method was established to qualitatively analyze four common components of snake bile,including Taurocholic acid(TCA),Taurochenodeoxycholic acid(TCDCA),Taurodeoxycholic acid(TDCA)and Cholic acid(CA),and providing the basis for the formulation of the quality standard of snake bile.Methods:1.A TLC identification method was established for the determination of snake bile by contrast with Taurocholic acid(TCA)and snake bile.Snake bile was quickly identified with other animal biles.2.Use polymerase chain reaction(PCR)method to identify the special properties of snake bile.3.UPLC-QTOF-MS was adopted to the snake bile qualitative analysis to find out the characteristics of the snake bile.Provide basis for quantitative analysis.4.HPLC-CAD method was established to qualitatively analyze four common components of snake bile,including Taurocholic acid(TCA),Taurochenodeoxycholic acid(TCDCA),Taurodeoxycholic acid(TDCA)and Cholic acid(CA).Results1.The solutions of different snake biles contained TCA.In pig bile,there was no TCA,cattle bile and sheep bile contained other spots besides TCA.There was a significant difference between snake bile and other animal biles.2.Compared with other analysis methods,Polymerase chain reaction(PCR)is more specific.3.The results showed that there were 5 common ingredients in the 12 batches of snake bile,and the differences in the composition of bile in different animals were significant.4.HPLC-CAD method was established to determine the known content of four common components in snake bile and the methodology was verified,the results showed that this method is simple,high sensitivity,suitable for quantitative analysis of snake bile.Conclusion:Through the above researches,this study can quickly identify the bile of 4different animals.The methods established in this experiment has the characteristics of simple operation,high sensitivity and high accuracy,which can be used as an important basis for quality evaluation of snake bile. |
