Background and Objective: Immunological thrombocytopenia(ITP)is a common clinical hemorrhagic hematological disease.In recent years,its pathogenesis is increasingly considered to be associated with cellular immune disorders.Studies have shown that in patients with ITP,the level of Th17 cells and the concentration of IL-17 in peripheral blood are significantly increased compared with the normal control group,suggesting that it is expected to become a new target for the treatment of ITP.As a new cellular immunotherapy,mesenchymal stem cell transplantation has been applied to the clinical treatment of many autoimmune diseases.Mesenchymal stem cells have immunosuppressive effect on Th17 cells and may be effective in the treatment of ITP,but there is no exact relevant report.We propose to establish a co-cultured cell model to study the role of mesenchymal stem cells in the differentiation of Th0 cells into Th17 cells.Methods Results:(1)Bone marrow-derived mesenchymal stem cells from BALB / c mice were isolated by adherence method.Morphological observation,plastic adherence,flow cytometry and immunofluorescence detection of cell surface markers(CD105 +> 85 %,CD90 +> 90%)are used to identify that they are mesenchymal stem cells.(2)Specific cytokines and antibodies induce the expression of a certain percentage of Th17 cells in spleen mononuclear cells,co-cultured with MSCs in a ratio of 20: 1,and the expression of Th17 cells(IL-17A)and Th1 cells(IFN-?)in the culture system was detected by flow cytometry,the expression of Th1 cells(IL-2,IL-12,IFN-?),Th2 cells(IL-4,IL-5,IL-6)and Th17 cells(IL-17A)in cells were detected by Real-time fluorescence quantitative PCR.Results: After co-culture,the results of flow assay and real-time fluorescence quantitative PCR indicated that the expression of Th17 cells and Th1 cells in the experimental group decreased significantly compared with the control group,and the difference was statistical significance.The results of real-time fluorescence quantitative PCR suggested that the expression of Th2 cells in the experimental group increased significantly compared with the control group,and the difference was statistical significance.Conclusions: The model of mesenchymal stem cells inhibiting the differentiation of Th0 cells into Th17 cells was successfully established.At the same time,it was found that mesenchymal stem cells inhibited the differentiation of Th0 cells into Th1 cells and had opposite effect on the differentiation of Th2 cells at the level of gene.
