Objective This study was designed to investigate the level of the plasma miRNA-183 and to discuss their changes in patients with non-small cell lung cancer(NSCLC)and to provide evidence for the diagnostic value of plasma miRNA-183 on NSCLC.Methods 1.Study Objects:The subjects in the study including 45 patients with NSCLC were chosen randomly,who were hospitalized in the Department of Cardiothoracic Surgery of Hunan Provincial People's Hospital from September 2017 to December 2017.Another 47 healthy subjects who were selected from Center of Health Examination were regarded as the control group.2.Experimental steps:Samples of peripheral venous blood were collected from the experimental subjects.Plasma was extracted after centrifugation.The reagent was used to extract plasma miRNAs.Then cDNA was synthesized by reverse transcription.Real-time fluorescence quantitative PCR(RT-qPCR)was used to detect the Ct value of miRNA-183 in plasma.?Ct value=Ct value of objective gene-Ct value reference gene.The relative expression level of plasma miRNA-183 was calculated by using the formula 2-??Ct.Meanwhile,peripheral venous blood samples were detected for level of carcinoembryonic antigen in Nuclear Medicine Department.3.Statistical Analysis:The statistical analysis of the data was based on the SPSS21.0 software.The difference of measurement data was compared by student's t-test between the two groups.The difference of enumeration data was compared by chi-square test.The difference was considered statistically significant when P<0.05.The difference of plasma miRNA-183 and CEA between the two groups was compared.The correlation of plasma miRNA-183 level with clinical and pathological features in NSCLC patients was analyzed.The author draw Receiver Operating Characteristic Curve(ROC)according the experimental data and calculated the area under curve(AUC)to compare the diagnosis value between plasma miRNA-183 and CEA in NSCLC.In the end,the author analyzed the diagnosis value in NSCLC by combining the plasma miRNA-183 and serum CEA.Result 1.The relative expression level of plasma miRNA-183 in experimental group and control group were(2.77±1.30)and(1.36±0.82)respectively.The difference was considered statistically significant(P<0.001).2.The relative expression level of serum CEA in experimental group and control group were 3.87(2.44-5.29)and 2.45(1.48-3.33)?g/L respectively,The difference was statistical significance(P<0.001).3.The relative expression level of plasma miRNA-183 in NSCLC patients was related to TNM staging,tumor diameter,lymph node metastasis and degree of differentiation(P<0.05),but not to gender,age,smoking,pathological type and pleural effusion(P>0.05).4.According to the receiver operating characteristic curve(ROC),the optimal cutoff value of plasma miRNA-183 relative expression to diagnose NSCLC is 2.13,with a sensitivity of 77.8%,specificity of 87.2%,AUC of 0.827.The optimal cutoff valrue of serum CEA expression to diagnose NSCLC is 3.32,with a sensitivity of 64.4%,specificity of 76.6%and AUC of 0.728.The combination of plasma miRNA-183 and serum CEA in the diagnosis of NSCLC show a sensitivity of 89.7%,specificity of 91.1%,AUC of 0.828.Conclusion 1.The level of plasma miRNA-183 of patients with NSCLC was up-regulated when compared to healthy group,which indicate it can be used as a potential molecular marker for the diagnosis of NSCLC.2.It can help to improve the diagnostic value of NSCLC by combining the plasma miRNA-183 and serum CEA.
