HNF4a,hepatocellular nuclear factor 4a,is the key transcriptional factor in liver development and homeostasis.Studies taken place in HCC had shown a decreased expression level of HNF4a in HCC comparing those of the normal tissue.Other studies had even explained the mechanism of the repressor role HNF4a played in HCC.Hippo pathway is one of the pathways controlling cell proliferation and differentiation of vital importance.The core component of Hippo pathway,YAP,is a typical proto-oncogene.YAP is overexpressed in many types of cancer,and has the characteristic of widespread especially in HCC,showing its relationship with HCC development.Our research group had done a lot of works between YAP-TEAD and HNF4a,and had found the double negative feedback loop between them.In this paper,we did more works using cell,rat and mouse model,with the technique of western blot,Real-time PCR,Luciferase,IHC etc,figuring out the suppressor role HNF4a played in mediating YAP-TEAD from different views.Due to the widespread effection of YAP in HCC,our research has shown a new way to future HCC treatment,by suppressing YAP-TEAD activity by HNF4a,and further more restrain HCC development,making it of vital importance for future HCC targeting treatment.
