For the purpose of switchable photoacoustic(PA)/fluorescence(FL)image,multiphase photothermal therapy(PTT)&photodynamic therapy(PDT),and synergetic theranostic of tumor,we developed zinc phthalocyanine-phospholipid(ZnPc-SPC)complex based drug carrier.In the first part,ZnPc-SPC complex was developed by a co-solvent method.NMR,XRD,FTIR,and UV-vis-NIR analysis demonstrated the weak molecular interaction between ZnPc and SPC.ZnPc-SPC self-assembled nanoparticle(ZS)was formed by a nanoprecipitaion technique.DSPE-PEG2000-MTX was introduced on the surface of ZS attaining ZSPM to endow them with folate receptor(FR)-targeting property.In the second part,we developed a zinc phthalocyanine-phospholipid complex based drug delivery system with Doxorubicin(Dox)as loading cargo to achieve additional chemotherapy while the carrier itself could serve as multifunctional and switchable theranostic agent.In the early phase,ZSPM and DZSM could display PTT and PA effect.Temeparature could reach as high as 43 ?.PA singnal reached maximum in tumor area at 2 h post-injection.While in the late phase,ZSPM and DZSM dis-assembled into ZnPc-SPC complex monomers again performing PDT and low-background FL image.ROS could be produced effectively.FL signal reached maximum in tumor area at 24 h post-injection and was stronger than in other organs.Besides,Dox was able to be released from DZSM achieving the synergetic therapy.In vitro and in vivo evaluations both indicated that ZSPM and DZSM presented high selectivity for FRa over-expressed tumor cells,significant cytotoxincity and anti-tumor effect in vivo.In a word,ZSPM and DZSM presented excellent switchable PA/FL image,significant multiphase PTT/PDT effect,as well as great synergetic therapy potential in DZSM,both serving as great candidates for multifunctional preicise tumor theranostics.
