Objective:to investigate the diagnostic value of Cyclin-dependent kinase 9?CDK9?for coronary atherosclerosis and its correlation with severity.Method:1.The correlation research between CDK9 and coronary atherosclerosis lesion:?1?.A total of 79 patients with concurrent coronary angiography were selected as the research object,which from the department of cardiology of the affiliated hospital of southwest medical university in January2017 to June 2017.According to the results of coronary angiography examination,patients can be divided into two groups:normal control group?no vascular stenosis?and coronary atherosclerosis group?vascular stenosis?which including vascular stenosis<50%of the patients included in the CHD group,vascular stenosis>50%of patients in coronary heart disease group.?2?.In the early morning fasting period,the patients were extracted and the serum concentration ofCDK9 and IL-6 were determined by enzyme-linked immunoassay?ELISA?.?3?.at the same time,the baseline data,blood lipid,liver function,myocardial enzyme spectra,cystatin C and B brain natriuretic peptide precursors were collected to analyze the differences of biochemical indexes.2.The correlation research between CDK9 and atherosclerotic lesion in rats,and to study Effect of the CDK9 inhibitor--Favopiridol on atherosclerotic lesions in rats:?1?.the same time SD male rats were randomly divided into group A?blank control group?,group B?group of atherosclerosis?and group C?Favopiridol treatment group?.Based feed in group A,group B and C group on the basis of a high-fat diet given by intraperitoneal injection of VitD3,FeSO4 join water preparation in the rat model of atherosclerosis,groups of rats feed 16 weeks.?2?.in the eighth week of feeding,group C gave a one-time intraperitoneal injection of Favopiridol to observe the effect of CDK9inhibitor on the degree of atherosclerotic lesions in rats,while the group B was given a one-time intraperitoneal injection of the same amount of normal saline.?3?.To collect three groups of rats serum at 8,12,16 weeks respectively,the serum concentration of CDK9 and IL-6 were determined by enzyme-linked immunoassay?ELISA?,and at the same time,testing groups of rats'serum concentration of total cholesterol?TC?,triglyceride?TG?,low density lipoprotein?LDL-C?,high density lipoprotein?HDL-C?in the clinical laboratory of first affiliated hospital of southwest medical university.Rats blood after death,collect each rat aortic blood vessels,and in the 16 weekend,to collect three rat groups'heart,liver,kidney and lung tissue,all pathological tissues were stained with HE staining.3.The pathological specimens of rats were compared with microscope and electron microscope.SPSS22.0 statistical software was used for significant?P<0.05?.?4?.CDK9predict coronary atherosclerosis area under the ROC curve?AUC?was 0.944?95%CI 0.898 to 0.991,P<0.001?,higher than troponin I?0.768,95%CI 0.667to 0.868,P<0.001?and b-type brain natriuretic peptide precursor?0.707,95%CI 0.594 to 0.820,P=0.003?.According to the CDK9 calculated by Youden index,the optimal boundary value of coronary atherosclerosis is 0.775,and the sensitivity of the prediction of coronary atherosclerosis is 81.5%,and the specificity is 96%.?5?.The area under ROC curve?AUC?predicted by CDK9was 0.863?95%CI,0.7690.958,P<0.001?,lower than troponin I?0.906,95%CI 0.8190.994,P<0.001?.According to the CDK9 calculated by Youden index,the optimal threshold of coronary atherosclerosis was 0.593,and the sensitivity of coronary atherosclerosis was 79.3%,and the specificity was80.0%.2.The correlation research between CDK9 and atherosclerotic lesion in rats,and to study Effect of the CDK9 inhibitor--Favopiridol on atherosclerotic lesions in rats:?1?.Rats in group A did not see atherosclerosis change at 16 weeks;The rats in group B showed mild atherosclerosis in the 8th week,and the longer the feeding time,the more obvious the atherosclerosis was,and the typical atherosclerosis was seen at the end of 16;The rats in group C had mild atherosclerosis in week 8,and the severity of atherosclerosis in 12 and 16 weeks was increased,but the typical atherosclerosis was not observed.?2?.Comparison in the group:the serum concentration of CDK9 and IL-6 showed no significant difference at the 8th,12th and 16th weeks in group A?P>0.05?.In group B,the serum concentration of CDK9 and IL-6 were gradually increased at weekend 8,12,16;and the differences were statistically significant in the 8th,12th and 16th weeks?P<0.01?.In group C,there was no significant difference between the 8th and 12th weeks?P>0.05?,and the concentration was higher than that at week 16,and the difference was statistically significant?P<0.01?.?3?Comparison between groups:on the 8th and 12th weekend,there was no statistical difference between groups A and C?P>0.05?.In group B,the level of CDK9 and IL-6 were increased compared with groups A and C,and the difference was statistically significant?P<0.01?.At the end of the 16th week,the levels of CDK9 and IL-6 in the three groups were gradually increased by group A,group C and group B,and the difference was statistically significant?P<0.01?.Conclusion:1.CDK9 is significantly increased in serum of patients with coronary atherosclerosis,and the more severe atherosclerosis is,the higher the expression concentration is.CDK9 has the ability to predict the occurrence of atherosclerosis.2.Early use of CDK9inhibitors in atherosclerosis on rats can delay the development of atherosclerosis.
